Nowadays, research on the physiological active substances from the marine tunicates is in a high speed progress. But it has not been reported that some extracts which have high bioactivity substances are obtained from The Styela clava. The research work carried on Styela clava in homeland is rarely reported. There are much many tuicates in our country, especially in Shandong province. These tuicates greatly disturbed the cultivation of water products. Therefore we should pay attention to them. In this paper, we made a basic research on bioactivity substances from Styela clava in following aspects state below.First, Styela clava'tunica and its body are separated carefully so that interaction among some substances is prevented. By using the traditional methods of extraction and isolation of natural products from Styela clava'body. The body was extracted with 100% chloroform and the solvent was separately concentrated under vacuum to yield a dark-brown gum. Then, we use the methyl alcohol re-solvent the gum and got methyl alcohol's extraction, this material was loaded onto a silica gel column and the column was eluted with chloroform/ methyl alcohol. Through this process, we got eleven extractions. Among these extractions, we found the F-2 was suited to the further research and load it onto silica gel column. Through this process, we got five extractions and found FG-2 and FG-4 were suited to the further research. By using the HSCCC to the two extractions, we got the Y-1 and Y-2, which were the high-purity.Second, using modern analytic techniques such as nucleic magnetic resonance (NMR), mass spectrometry (MS), GC-MS, HPLC-MS, we analyzed the basic chemical structure of Y-1and Y-2. Results showed that Y-1 was ethoxylated fatty alcohol and Y-2 was Phthalic acid, bis-iso-butyl ester.At last, we investigated the Y-1 and Y-2'anticancer activity by culturing cancer cells such as HepG2 liver cancer cell and HO8910pm ovarian cancer cell, and the antiproliferative effects were measured by MTT assay. From the data of the experiment we found that we found that the vitro anti-tumor activities of Y-1 and Y-2 was instable, no regular concentration and time dependence. For the Hepg2 liver cancer cell, the effect of Y-1 and Y-2 with the different time and concentration is very mild. For the HO8910pm ovarian cancer cell, the maximum inhibition rate of Y-1 was 22.94 % under the concentration 100μg/mL after 24h of incubation and this effect is not time and concentra- tion-dependent The maximum inhibition rate of Y-2 was 17.28 % under the concentration 100μg/mL after 24h of incubation. But after the 48h of incubation, the inhibition activity became weaken.Although in the paper we didn't receive satisfactory results, this research is a certain innovation in the field of domestic work. Basing on this paper, further extensive screening, chemical and biological activities study should be developed, in order to find the novel bioactive substances to the development of a new drug or drug design as a precursor compounds. It can also provide valuable tool medicine for the basis life science research. We believe that further experimental studies can push forward the domestic work in the field of natural products research and development, and start the new situation of ascidians physiology active material studies.
