Research On The Structure Elucidation And Bioactivity Of The Active Substances From Styela Clava

Nowadays, research on the physiological active substances from the marine tunicates is in a high speed progress. But it is still not seen on papers that some tunicate extracts which have high effective anticancer potential are obtained from Styela clava's tunica and body or other kinds of substances. And the research work carried on Styela clava in homeland is rarely reported. Our country especially coastal areas in Shandong province abounds tunicates, But comparing with the overseas, the resources greatly disturbed the cultivation and sailing, not mention they are utilized. Therefore we should pay attention to them. In this paper, we made a basic research on the physiological active substances from Styela clava in following aspects stated below.First, Styela clava's tunica and its body are separated carefully so that interaction among some substances are prevented. In this paper, we focus only on the chemical compoud from the body of Styela clava. According to the frequently used methods of extraction and isolation of natural products from organic body, the freeze-dried ascidian body was exhaustively extracted with methanol/toluene ( 3 : 1 ) and then the combined extracts were filtered and the solvent was separatelyconcentrated under vacuum. The concentrate then was partitioned with IN NaNOs , The aqueous phase was collected and extracted with chloroform .Then the chloroform extact was concentrated under vacuum to yield a red-brown gum. This material was loaded onto a silica gel column and the column was eluted with chloroform several times untile the fraction of interest was collected. Finally we obtained some light yellow ointment liquid which was a admixture and was named F I .Second, Method of GS-MS was used to analyze components of F I . Results showed that the main component of F I is l,2-bis(5-methyl-2 -benzoxazol)-ethene.At last,we investigated the F I 'anticancer activity by culturing cancer cells such as BEL7402 liver cancer cell and BGC823 gastric cancer cell, and antiproliferative effects were measured by MTT assay. From the data of the experiment we found that F I can inhibit the activity of BEL7402 liver cancer cell at different time and different dosage. The remarkable effect happened after 48h of incubation with F I under the concentration 60u,g/ml. F I can inhibit the activity of BGC823 gastric cancer cell significantly only under the concentration 60u.g/ml. In addition, we investigated the F I 'activity of immune-mediate by culturing the spleen cells of mice. All the results implied that the F I has not any remarkable effects on proliferation of lymphocyte and production of cytokines such as IL-2 and IL-6.We believe that through further research, the work in this field in our country will make a rapid progress and a branded-new prospect will come out.
