Objective To observe the time of adenovirus vector mediated rat insulin-like growth factor-1 (rIGF-1) gene expression in male Wistar mice. And observe the difference among different ways of injection.Methods 293 cell was used to culture adenovirus. One hundred and eighty male Wistar mice aged from four to five weeks were randomly divided into five groups. Group 1 is normal control group without special disposal; Group 2 received the intraperitoneal injection of adenovirus vector mediated empty green fluorescence protein (Ad-eGFP). Group 3 to 5 received injection of adenovirus vector mediated rIGF-1 gene (0.1ml) separately by intraperitoneal injection, intramuscular injection and pancreas tegument injection. Each group was randomly divided into six small groups. Mice were fed and libitum with a standard diet. The six small groups would be executed one by one in the third, seventh, fourteenth, twenty first, twenty eighth and thirty fifth day after disposal. All mice were executed and pancreata were fixed for more 12-24 hours, embedded in paraffin, and sectioned. We observed the expression of rIGF-1 detected with the immunohistochemical, enzyme linked immunosorbent assay (ELISA) techniques and reverse transcriptase polymerase chain reaction (RT-PCR). The plasma concentration of insulin was detected with enzyme linked immunosorbent assay (ELISA) techniques. Weight and glycemia of the sixth small group mice were measured once a week.Results Compare with control group, immunohistochemical and reverse transcriptase polymerase chain reaction show that experimental group have a higher rIGF-1 expression in islet cell from the seventh day to the twenty first day after injection; at the same time, rIGF-1 in serum detected by ELISA show a higher degree in experimental groups. There was no difference in different injection ways. The body weight of mice in experimental groups increased higher than control group accompany with the expression of rIGF-1 gene. The plasma concentrations of insulin detected with ELISA techniques have no difference in all groups. No hypoglycemia manifestation was found during the experiment.Conclusion These results suggested that adenovirus vector mediated rat insulin-like growth factor 1 (rIGF-1) gene have high expression in male Wistar mice's body from the seventh day to the twenty first day after injection. The results were no difference among different injection ways. IGF-1 improves Wistar mice's body weight and has no influence in blood glucose and the plasma concentrations of insulin.
