Objective:To investigate the relationship between a functional single nucleotide polymorphisms(SNP) in the core promoter region of GDF5 gene(+104T>c; rs143383) and spinal fusion in Qingdao Han people.Methods:Genomic DNA was extracted from the venous blood and identify the SNP of GDF5 gene(+104T>c; rs143383), using PCR-restriction fragment length polymorphism(RFLP). X-ray examinations were done after operation at 3,6,12 months in all the patients. Evaluate the degree of bony fusion according to the lateral and dynamic view and analyze the correlation between SNP and spinal fusion.Results:In all the 401 cases, there are+104T>C SNP in both the case group and the control group. There is no statistic significant difference between the case group and the control group. In the fusion group,57 cases are TT genotype, and 100 cases are TC+CC genotype. There are 24cases with the TT genotype and 20 cases with TC+CC. The difference between the two groups is statistic significant (p=0.023). In the fusion group, the fast case was 52, medium was 34, slow was 14, with the TC+CC genotype. It shows significant with the TT genotype (p=0.007).Conclusions:SNP in the core promoter region of GDF5(+104T>C) is associated with spinal fusion in Qingdao Han people. The allele C may be an important factor to promote spinal fusion.
