ObjectiveThe intratypic sequence mutation of HPV16 are known to differ in geographic distribution. The aim of this study was to investigate the distribution of HPV16 E6 and E2 gene mutation in cervical cancer to identify the most prevalent and novel HPV16 mutations in Qingdao, Shandong.Materials and Methods[1]104 cervical cancer cases consisted of 43 fresh tissues and 61 paraffinaceous tissues were recruited, including 95 squamous carcinoma,6 adencarcinoma and 3 adenosquamous carcinoma.[2] DNA samples were extracted from the cervical cancer tissues.[3]DNA samples were amplified by polymerase chain reaction(PCR) using specific primers for high-risk HPV, HPV16 to identify the positive tissues.[4] DNA samples extracted from the HPV16 positive tissues were amplified by the primers for E6 and E2.[5]Then the PCR fragments of HPV16 E6 and E2 genes were purified, sequenced and compared with the type stain.Results[1]The positive rate of HPV and HPV16 was 93.27%(97/104) and 69.23%(72/104) respectively in Qingdao, Shandong.[2] Among 104 cervical cancer tissues,37 E6,11 E2X and 23 E2Ywere amplified and sequenced successfully, for the efficiency of amplification low in paraffinaceous tissues. Prototype HPV16 E6 was found in 5 cases(13.51%)while mutation was detected in 32 cases (86.49%),among which the T178G or T178A (D25E) mutation was found in 25 cases(67.57%).The E2 sequence of 23 cases contained C3684A(T-K), that of 14 cases contained T3524C,C3684A (T-K) and C3787A (D-E) at the same sample, and 8 cases contained A2926G, C3159A (T-K), G3249A (R-Q), T3384C (I-T),C3410T (P-S), C3684A(T-K) and C3787A(D-E) meanwhile.Conclusions[1]The infection rate of HPV is close to the level of the world in cervical cancer in Qingdao.[2] There are several HPV16 E6 and E2 gene mutations in cervical cancer tissue of Qingdao women.
