Abstract/Summary:
BackgroundMyocardial infarction and other cardiovascular diseases are common causes of cardiac failure, and the irreversible necrosis of myocardial cells is one of the most important aggravating factors of cardiac failure. Traditional therapies such as drugs, intervention or surgery can not make necrosis cells regenerate. Recent studies revealed that endothelial progenitor cell was a kind of precursor cell which can differentiate into vascular endothelial cell directly and played an important part in the angiogenesis and the repair of ischemia tissues, which can improve the blood-supply of the cardiac muscle and the cardiac function after the myocardial infarction. Based on this theory, we transplanted the endothelial progenitor cells derived from bone marrow into the ischemia cardiac tissues and observed the cardiac function and plasma BNP level in rabbits with myocardial infarction.ObjectiveTo explore the effect of endothelial progenitor cells transplantation on cardiac function and plasma BNP level in rabbits with myocardial infarctionMethodsPart 1 Culture, identification and BrdU tagging of rabbits EPCs derived from bone marrowMonocytes derivded from the rabbit bone marrow via density gradient centrifugation were cultured in the EBM-2 medium and observed regularly. The EPCs were identified by expression Flk-1 and AC133. The EPCs were labeled with BrdU before transplantation.Part 2Myocardial infarction was induced by ligation left anterior descending branch of coronary artery. Then, the rabbits models were treated with aspirin, clopidogrel, isosorbide mononitrate capsules, atorvastatin calcium, angiotensin converting enzyme inhibitor,β-adrenergic blocking agent(doses were determined accroding to standard body weight) and randomly divided into three groups: the EPC group, vehicle group and sham group. In the vehicle group, PBS was injected into the cardiac muscles, while the PBS with endothelial progenitor cells was injected into the cardiac muscles in the treatment group. 4 weeks after the injection, echocardiography, capillary density in infracted area and BNP level were tested to measure the cardiac function.Results1. The cells derived from bone marrow are Flk-1 and AC133 positive cells.2. Compared with vehicle group, the LVEF of EPC group was increased(P<0.05). But the LVEDd of EPC group had no significant difference from that of vehicle group.3. The capillary density in infracted area in the EPC group significantly increased compared with vehicle group(P<0.01).4. The Plasma BNP level of EPC group decreased significantly, compared with vehicle group (P<0.05).ConclusionEndothelial progenitor cells transplantation can improve cardiac function and decrease the level of plasma BNP in the rabbits wih myocardial infarction.
