Objective: To study the inflectional situation of ORF26 subtypes of Human herpesvirus-8 in squamous cell carcinoma (SCC) ?mycosis fungoides (MF) ?Castleman's disease (CD) ?pyogenic granuloma (PG) in Xinjiang, and to discuss the associations between HHV-8 and different kinds of tissues. Methods: HHV-8 DNA was extracted with traditional phenol-chloroform-isoamyl alcohol from 30 cases SCC?13 cases MF?20 cases CD and 20 cases PG formalin-fixed, paraffin-embedded tissue from the department of dermatologists and pathology of People's Hospital of Xinjiang Uygur Autonomous Region from 2004 to 2009. KS is used as controls. The extracted DNA of ORF26 of HHV-8 was amplified by nested PCR. Then PCR product was electrophoresised at the 1. 5% agarose gel and ORF26 gene positive samples were observed under the gel imaging system. The analysis of positive specimens of ORF26 gene was carried out . Results: HHV-8 DNA was all detected in 5 cases SCC (16. 7%) and in 1 cases MF (7. 7%) . There was not HHV-8 in the tissues of PG and CD. Conclusion:â‘ HHV-8 infection in SCC of skin has higher rate, and that may be the cause of SCC of skin in XinJiang.â‘¡HHV-8 infection in MF may be one kinds of secondary infection . NO evidence of HHV-8 infection was found in others two diseases.
