Objective: Fracture is a kind of severe trauma. With the developments of transportation, there are more and more trauma patients nowadays. It is a dream of each orthopedist to accelerate fracture healing and to reduce healing time. It is a hot topic for researchers on accelerating fracture healing at home or overseas. Although many ways to accelerate fracture healing exists, including biological method, physical method, there is no satisfactory treatment modality at present. Chinese medicine has a history of thousands of years. During the long practice period, many Chinese prescriptions have been concluded and applied to healing fracture, and"jie gu yi hao"is one of the typical Chinese prescriptions. Both experimental research and clinical research demonstrate that Chinese medicine has a positive effect on fracture healing, especially accelerating it, however, the interaction mechanisms are not so clear. Many experimental researches prove that Chinese medicine can accelerate fracture healing through the aspects of accelerating blood circulation ,improving absoption of hematoma and organization, increasing collagen synthesis, raising numbers of osteoblast, increasing cell activity ,adding calcium salts deposit, improving poroma quality, and so on .There is an ideal method to clarify that Chinese medicine could improve fracture healing .The method , using modern scientific theory and technology, is appropriate to deep into the levels of cell and molecule ,for the time of researches of fracture is the era of molecular biophysics. In the experiment, we used the experimental medel of Newsland rabbit radius-defect,and investigated the variation of BMP-7 before and after Chinese medicine treatments. BMP-7 mRNA expression were measured by RT-PCR, and BMP-7 proteins expression were measured by immunocytochemistry. Through the levels of mRNA and protein, we tried to know and to find the evidence of the fracture healing mechanism of Chinese medicine.Methods: 40 healthy mature Newsland rabbits were randomly assigned into 2 groups, one experimental group and one control group. The 3mm-long bone defects were made in both radii of each rabbit. The experimental group was treated by"jie gu yi hao"external application, while the control group was not treated. After 2w,4w,6w,sacrifice the experimental animal respectively. Then, assess the fracture healing through X-ray and HE staining. After 2w,3w,4w,5w,6w,observe the changes of gene and protein expression of BMP-7 through immunohistochemistry and RT-PCR in the course of treatment.Results:X-ray shows: experimental group's bone callus appeared earlier and having more quantity bone callus experimental group's fibronectin and osteoblast occurrs earlier than control group through HE stain; the experimental group aslo has more callus. Immunohistochemical study group IOD (optical density value). Finally, do statistical analysis as the mean comparison of two samples't test into gray value of RT-PCR and MOD of immunohistochemistry. The results show that the difference is statistically significant.Conclusion:"Jie gu yi hao"can promote fracture healing by improving blood circulation, promoting hematoma absorption and machines, and promoting calcium deposition, improving the quality of callus and level of the expression of bone morphogenetic protein -7 (BMP-7).
