Objective:To study the distribution of allelic genotypes of single nucleotide polymorphisms of Klotho G-395A, F352V, C370S and to research the relationship of Klotho gene polymorphisms with heart rate variability in elderly patients with essential hypertension in the Chinese Han nationality population.Methods:Unrelated 114 elderly hypertension patients and 42 healthy individuals were chosen and genomic DNA of leukocytes was extracted from venous blood. Allele-specific PCR technique was used to detect genotypes of single nucleotide polymorphisms (SNP) at the sites of G-395A, F352V, C370S of Klotho gene.The heart rate variability (HRV) parameters such as time domain and power spectral components were analyzed.Results:Allele frequencies of GA+AA genotypes of Klotho G-395A polymorphism in elderly hypertension patients(60.18%)was higher than that in control group(40.48%)(P<0.05); There was no significant difference in frequency of all genotypes and allelic genes of both Klotho C370S and Klotho F352V(P>0.05).There was no obvious difference in the related parameters of HRV compared to each other in those with GG,GA and AA allele genotypes of Klotho G-395A polymorphism in elderly hypertension patients (P>0.05). There was no significant difference in the related parameters of HRV compared to each other in those with FF,FV and VV allele genotypes of Klotho F352V polymorphism in elderly hypertension patients (P > 0.05).The measured parameters of HRV, including TP, ULP, PNN50, RMSSD, and SDNN index, in the elderly patients with essential hypertension who carried with CS genotype were significantly higher than those in the patients with the CC genotypes of Klotho C370S (P<0.05).There was no difference in the related parameters of HRV between those with CS and SS allele genotypes of klotho C370S polymorphism in elderly hypertension patients (P>0.05).Conclusions:We observed that people with GA/AA genotype of G-395A SNP of Klotho gene were liable to suffer from essential hypertension. The related parameters of HRV were correlated with Klotho C370S polymorphism, suggesting that elderly patients with essential hypertension who carry with CS genotype of Klotho C370S be at high risk of cardiac autonomic nerve function disturbances. Meanwhile there is no association between the related parameters of HRV and KL G-395A or F352V SNP.
