Preliminary Study Of The Effect Of Low-intensity Pulsed Ultrasound United GBR Technique (bio-gide Collagen Membrane) On Promoting Fenestration Repair In BEAGLE Dogs' Canine Periodontium

Low Intensity Pulsed Ultrasound (LIPUS) as a non-invasive mechanical energy and safe in theory has the potential in repairing the periodontium with non-invasive.So far, there were no reports about the model about LIPUS united Guided Bone Regeneration (GBR) technique to repair fenestration in periodontium.In this study, we established the peridontal fenestration model of Beagle dogs'canine by periodontal surgery. To investigate the repair effect of model about Beagle dogs'canine fenestration in periodontium, we also made the model combined with Autogenous bone graft, GBR and supplemented LIPUS irradiation.Objects of this study were to set up a 5×5mm2 acute bone defect model of peridontal fenestration in the canines' buccal alveolar area of 8 adult male Beagle dogs.The four canines of each Beagle dog were randomly divided into 4 groups.Those were control group, Bio-Gide collagen membrane+autogenous bone graft group, Bio-Gide collagen membrane+autogenous bone graft+LIPUS group and autogenous bone graft+LIPUS group. The experimental groups that involved LIPUS would be Irradiated 1 per day for 20min (Ultrasonic intensity through the collagen membrane 30mw/cm2 , frequency of 1MHz ,pulse width modulation signal 200μs, repetition rate of 1KHz ) Ultrasound treated for 6 and 8weeks,animals were sacrificed to do the correlation detection about the hard tissue of experiment area and Micro-CT.In the group of autogenous bone graft+LIPUS and Bio-Gide collagen membrane+autogenous bone graft+LIPUS,the slice of hard tissue showed that osteoblasts and cementoblasts distributed intensive, there were lots of regenerate collagen fiberⅡ.That indicated that the alveolar bone had a higher maturity. In the group of Bio-Gide collagen membrane+autogenous bone graft group and Bio-Gide collagen membrane+autogenous bone graft+LIPUS group, the number of regenerate peridontal cells and the intensity of regenerate collagen fiber were better than the other groups. But the difference between them were still difficult to analyze clearly by dimensional image.BV,BS/BV,BS,Tb.N,Tb.Sp,Tb.Th had been detected by Micro-CT.In the 6th week,there were significant differences between BVand BS(P<0.05). In a certain time basis,there were significant differences among BV,BS,Tb.Nand Tb.Sp((P<0.05).But between 6th and 8th week,there were no significant differences(P﹥0.05).This all prompted that LIPUS united GBR technique play a positive role on the Beagle dog canine periodontal "fenestration"model. In addition ,GBR technique play a certain role in inhibiting bacterial invasion.LIPUS had the potential to promote peridontal cells to generate and differentiate.So combining both the effect could be reduced on the one hand,on the other hand the model of fenestration repair in periodontium had been enhanced.In sum, the study found ,to some extent,certain parameters LIPUS (Ultrasonic intensity through the collagen membrane 30mw/cm~2 , frequency of 1MHz ,pulse width modulation signal 200μs, repetition rate of 1KHz ) could promote the remodeling and maturation of the Beagle dog canine's periodontium and alveolar bone structure .Moreover,it would obtain a better result if united GBR technique.For that,it has provided a new treatment for the treatment of periodontal tissue.however, further study is needed.
