Objective: To observe the effect of insulin (INS) on rabbit corneal alkali burn in the corneal tissue pathological changes and interleukin expression, and to explore its early anti-inflammatory mechanisms.Methods: 45 female New Zeal white rabbits were randomly divided into four groups: A group (saline group, n=15) , B group(dexamethasone group, n=15) and C group(INS group, n=15), to establishⅡdegree model of corneal alkali burn, and respectively to inject normal saline, dexameth- asone and ISN in the subconjunction each day(dose=0.075 ml/kg) until the end of experiment. Concentration of IL-1βwere detected by ELISA in blood and aqueous humor, and content of IL-10 mRNA were detected by rt-PCR in corneal tissue, and corneal morphological changes were observed in corneal pathological section, at 1 d, 3 d, 7 d.Results: In the central region of corneal alkali burn at 7 d: stromal fiber was still edema, and stromal cells were still missing at the A group; but corneal epithelium was thickening, edema of stromal fiber was mainly subsided, and a small amount of stromal cells were appeared in the groundmass at the B group and C group. it shows that C group and B group were better than A group about corneal recovery. After corneal alkali burn,the levels of inflammatory cytokines IL-1βincreased significantly in blood at 1 d, but at 3 d and 7 d decreased; B group and C group were significantly less than A group (P<0.01) about the levels of inflammatory cytokines IL-1βin blood and aqueous humor, at 3 d and 7 d. Compared with the control group (A group), the B and C group showed IL-10mRNA expression increasing as time increasing in corneal alkali burn tissue at 1 d, 3 d and 7 d. And the IL-10mRNA expression in both B and C group were significantly higher than A group(P<0.01).Conclusion: Early corneal alkali burn, INS can reduce pro-infla- mmatory factors and improve the content of anti-inflammatory factors, to play an anti-inflammatory role in promoting the restoration of corneal injury.
