ObjectiveCorneal neovascularization,(CNV) was common in corneal diseases,which was caused by many reasons.It is a major complication of corneal injury from chemical burns from accidental exposure to solvents,detergents and alkali,and of corneal infections including bacterial,Chlamydial,parasitic and viral infections Corneal vascularisation is a potentially devastating occurrence that can cause blindness. Treatment of neovascularisation of the cornea remains difficult with few effective specific therapies available.Current options include the use of topical corticosteroids which do not specifically inhibit angiogenesis,but have wide-ranging anti-inflammatory effects and are contraindicated for conditions where there is a risk of infection.After the cornea has become vascularised,grafting is "high-risk". Neovascularized cornea wound be rejected for the reason that the normal corneal structure was changed.And the 2-year survival rate of the graft dropping from approximately 90%to below 50%.The development of study for corneal vascularisation is therefore of great clinical significance.In this study,we observe the expression of CD105 in neovascularized cornea of rats after alkali burn which could provid some proofs for pathogenesy.Methods1.60 Wistar rats were separated to 2 groups randomly.In the experiment group, CNV was induced by alkaline cauterization of the central cornea.the control grope were receiced 0.9%Nacl to take the place of alkali.We give antibiotic eye drops to both groups after the animal model were established.2.After the animal model were established,we observed the length and the area of corneal NV by using slitlamp,at the same time took pictures of the eyes.3.Rats were killed at 1,3,5,7,10,14 d after alkali burn.corneal tissue wound be in the detection of pathology and the expression of CD105 was investigated with immnuohistochemical method in rat cornea immnuohistochemical method.Results1.The CNV was found from 2 days after alkali burn in the rat cornea.The CNV area peaked at 7 days and decreased from 10 to 14 days.2.HE dyeing showed that:in contral group the corneal tissue were well-arranged. At the 3~7 d,in the experiment group there were many CNV and red cells,at the same time the corneal epithelium were edema and the stroma were irregularity.We also found many inflammatory cell in cornea.When 10~14 d the CNV reduced and the degree of edema were to ease.3.In contral group the expression of CD105 were less.But the expression of CD105 increased in the experiment group at 3~7 d after cautery.CD105 protein peaked at 7 days after cautery,decreased after 7 days,and significant decreased after 14 days.There were statistic difference in the expression of CD105 between the two groups.ConclusionsThis study suggests that CD105 express significantly in neovascularized cornea of rats after rats' cornea were alkaline burned.It may participate the formation of the CNV.
