Objective:Study the status of promoter methylation of DLEC1 in non-small cell lung cancer and the association with its protein expression to explore the role of DLEC1 in lung cancer occurrence and development and to explore the association between DLEC1 and Ki-67.Methods:Use the method of methylation-specific PCR to detect the levels of promoter methylation of DLEC1 gene and use the method of immunohistochemistry assay ElivisionTM plus to detect the expressions of DLEC1 protein and Ki-67 antigen in non-small cell lung cancer tissues.Results:1. The rate of DLEC1 gene promoter methylation in 83 cases of non-small cell lung cancer tissues is 51.81%(43/83) and the rate of the methylation in the adjacent lung tissues is 7.23%(6/83),the difference was statistically significant (P <0.05);The rate of promoter methylation of DLEC1 of the patients during TNM I and II is 39.29%(22/56),and during TNM III andâ…£it is 77.78%(21/27),the difference was statistically significant (p <0.05).Patients with lymph node metastasis,the rate of promoter methylation of DLEC1 of them is 78.13%(25/32),and others without lymph node metastasis,the rate of promoter methylation of DLEC1 of them is 35.29%(18/51),the difference was statistically significant (P <0.05).Patients with squamous cell lung carcinoma,the rate of DLEC1 promoter methylation of them is 60.71%(34/56),and those who with adenocarcinoma of the lung,the rate of DLEC1 promoter methylation is 31.82%(7/22),the difference was statistically significant (P <0.05).DLEC1 gene promoter methylation has nothing to do with the age, sex, location, tumor size and level of cellular differentiation.2. DLEC1 protein expression rate of 83 cases of non-small cell lung cancer tissues is 25.30%(21/83),and in adjacent lung tissues,DLEC1 protein expression rate is 67.47%(56/83),the difference was statistically significant(P<0.05).DLEC1 protein expression has something certain to do with histological type,lymph node metastasis,TNM staging (P<0.05),but has nothing to do with the age,sex,location,size of tumor and level of cellular differentiation (P> 0.05). 3. Ki-67 positive expression rate of 83 cases of non-small cell lung cancer tissues is 78.31%(65/83),and in adjacent lung tissues,Ki-67 positive expression rate is 25.76%(17/83),the difference was statistically significant(P<0.05).Ki-67 antigen expression was significantly different from histological type, level of cellular differentiation,lymph node metastasis and TNM staging (P<0.05), but has nothing to do with age,sex,location, and tumor size (P>0.05).4. 9 cases of Ki-67 expression in 21 cases of DLEC1 protein positive expression were positive(42.86%),56 cases of Ki-67 expression in 62 cases of DLEC1 protein negative expression were positive(90.32%),according to the Spearmans correlation analysis,r=- 0.501, P<0.01,it indicates that in non-small cell lung cancer tissues DLEC1 and Ki-67 protein expression is negatively correlated.5. 3 cases of DLEC1 expression in 43 cases of DLEC1 gene promoter methylation in non-small cell lung cancer tissues were positive(6.98%), 15 cases of DLEC1 expression in 40 cases of non-DLEC1 gene promoter methylation in non-small cell lung cancer tissues were positive(45.00%), According to the Spearman correlation analysis, r =- 0.437, P <0.001, it explains that in non-small cell lung cancer tissues DLEC1 gene promoter methylation and DLEC1 protein expression is a negative correlation.Conclusions:1. DLEC1 protein with low expression in non-small cell lung cancer tissues is concerned with tumor histological type, lymph node metastasis, TNM staging,it can suggest that loss of expression DLEC1 and the occurrence plus the development of lung cancer is closely related.The existence of differences between DLEC1 protein in lung tumors and adjacent tissues can suggest that it may provide new clues and new ideas for the study of the diagnosis of tumors.2. The rate of DLEC1 promoter methylation in Non-small cell lung cancer tissues is significantly higher than that of the adjacent lung tissues,and was begatively correlated with DLEC1 protein expression,suggesting that promoter methylation may be the main way making DLEC1 the main form of gene silencing; in addition,the detection of DLEC1 promoter methylation may be of some significance for the auxiliary diagnosis of lung cancer.3. Expressions of DLEC1 protein and Ki-67 were negatively correlated, suggesting that DLEC1 genes may be involved in cell cycle control and inhibit the proliferation of tumor cell, which able to provide certain theoretical basis for making definite the functions of DLEC1 proteins.
