BackgroudLung cancer is one of the most common malignant tumor and the main cause of death in people,recent years, the morbidity and mortality of lung cancer is highest among the world’s malignant tumor.There are more than1600000new cases of Lung cancer each year in the word, and more than1300000persons are death of lung cancer each year. Lung cancer is mainly divided into small cell lung cancer(SCLC) and non-small cell lung cancer (NSCLC) according to its different histologic expression andbiological behavior. NSCLC accounts for about80%85%of all lung cancer. nowdays, scholars havebeen explored the mechanism of lung cancer from different angles. Cancer cells are different from normalcells in stronger prolification, less apoptosis, higher gene mutation rate, and the increased multi-drugresistance. This kind of phenomenon is associated with the expression of tumor markers in cancer cells,such as carcinoembryonic antigen (CEA), neuron specific enolization enzyme (NSE), squamous cellcarcinoma antigen (SCC Ag),selenium binding protein1(SBP1), etc. The process of which is verycomplex and also is not clear. In view of the high incidence of NSCLC, and this desease is found mostly inthe advanced grade, the prognosis of lung cancer is poor. Single tumor markers are not sensitive to discoverlung cancer. Exploring novel marker makes the main direction of further research of lung cancer.Selenium binding protein1(SBP1) is one of selenium-binding protein in the body, located inchromosome1q21-22, its mRNA sequence (Gene bank seat number: NM003944) consists of1721nucleotides encoding640amino acids, the molecular weight of it is56kda. Due to the human gene for SBPand mouse SP56is homologue, SBP1is also called hSP561. Domestic and international studies haveshown that with the increased intaking of selenium into the human body, the expression of seleniumbinding protein1(SBP1) is improved and the body’s resistance to cancer is also increased. Studies showedSBP1is lower expressed in many malignancies, which indicate it could be a maker in diagnosis of tumorand assessing the clinical prognosis of malignancy.Objective:Western blotting, immunohistochemical and rt–PCR were used to test the expression of SBP1in llung squamous carcinoma tissues, para-carcinoma tissues, normal tissues and benign tumor tissue through which to study the expression and significance of SBP1in tumorigenesis and development of lungsquamous carcinoma.Methods:This study is on surgical removal lung tissues which were derived from in hospital Pulmonary diseasepatients.20of them are lung squamous carcinoma cases,20are para-carcinoma cases,20are bemign tumorcases,total number is60. They were gathered into different groups according to gender, age, pathologicaltype and TNM staging. Western blotting, immunohistochemical and rt–PCR were used to test theexpression of SBP1in lung squamous carcinoma tissues, para-carcinoma tissues, and benign tumor tissues.Date was deal with by SPSS13.0.Result:1, Immunohistochemical staining showed that SBP1protein have been expressed in the cytoplasm andnucleus of the tumor cells. SBP1expression is lower in lung squamous carcinoma (4/20,20%), theexpression of it in para-carcinoma is14/20(70%), in the lung benign tumor is18/20(90%). The expressionof SBP1between lung squamous carcinoma and all other2type tissues is statistically significant (P <0.05).And the expression of SBP1among the other2types is of no statistically significance (P>0.05)2Western blot analysis showed that the expression of SBP1in benign tumor is more than that in lungsquamous carcinoma and the difference was significantly (P <0.05).3Rt-PCR showed that the expression of SBP1in lung squamous carcinoma tissues is lower than that in thepara-carcinoma tissues and benign tumor tissues.4.In lung squamous carcinoma the expression of SBP1is unassociated with age and histological grade(P>0.05); the expression of SBP1is associated with tumor size, lymph node metastasis and TMN clinicalstaging,and the difference was statistically significant (P <0.05)5.single-factor Cox stepwise regression analysis of20lung squamous carcinoma patients showed thatlymph node metastasis, TNM stage, histology grading and SBP1expression are factors affecting thesurvival of patients with lung cancer (P <0.05). Multivariate COX gradually back Classified analysisdisplayed that the TNM Staging (RR:1.862) and SBP1expression (RR:0.213)(P <0.05)is associated theprognosis of patients with lung squamous carcinoma.Conclusion: 1.The difference of SBP1protein expression in benign pulmonary lesion and lung squamouscarcinoma tissue expression was statistically significant; the expression of SBP1is unassociated with ageand histological grade but is associated with tumor size, lymph node metastasis and TMN clinical staging2. Multivariate COX gradually back Classified analysis displayed that the TNM Staging and SBP1expression is associated the prognosis of patients with lung squamous carcinoma r.3SBP1may become a index of the clinical diagnosis and prognosis judgment of lung squamouscarcinoma.
