Gene Tempral-spacial Expression Of Pulmonray Fibrosis And Intervention Study With Total Astraglosideo

Objective: Using rat gene chip to screen abnormal genes expression of pulmonary fibrosis and intervention study with total astraglosideo in different time-points.To elucidate the pathogenic mechanism of pulmonary fibrosis and related taget gene of drug action .Methods: the lung specimens of model group in 7d and 28d and total astraglosideo-intervented group in 28d were hybridizated with agilent 46000+ whole genome oligo microarray , the image scanning and date analysis were performed after hybridization . The reliability of the microarray result was confirmed by Real-Time PCR technology .Result: model-28d compare with model-7d,there are 2063 diferential espression genes ,we select 109 genes ,there are 43 genes up-regulated ,66 genes down-regulated.model-28d compare with model-A1 28d ,There are 4269 differential expression genes ,We select 68 genes , 45 genes are up-regulated , Rest of it are down-regulated. Using Go analysis and PATHEWAY analysis ,It shows that the genes selected involve in various aspects in form and development phase of pulmonary fibrosis. Picking up 3 genes ,Timp1,Ltbp2,Fgf18 ,to prove the validity of genechip result by Real-time PCR ,The result show all these 3 genes abnormally expressed in 3 samples .Conclusion : (1) the diferetial expression genes of pulmonary fibrosis induced by bleomycin in 7 days almost display phlegmanosis and fibrosis genes upgratulation..while these genes down-regulated in 28 days.(2)combine togethe pathological section and gene chip result , total astraglosideo has obviously therapeutical effect .the principal mechanism is to decrease inflammatory factor and fibrosis genes expression..(3) Selecting 3 genes , Timp1(tissue inhibitor of metallopeptidase-1),Fgf18 (fibroblast growth factor 18),Ltbp2(transforming growth factor beta binding protein 2),to carry out Real-time PCR test , the result is same as the gene chip .moreover the express capacity significant correlate with ratio value in genechip , it shows that it's ture and reliable to screen the differential genes of pulmonary fibrosis by genechip .
