Objective:Combining the rat's caudal vein of MCAO and that of its transplanting MSCs and EPCs, between which and single transplantation, a comparative study is made.Observing the effect different ways of transplantation made on cerebral ischemia injury of the rat, after which comparing the differences between neuroethology, cerebral infarct volume and pathomorphology, this paper probes into which way of transplantation is the Most Effective way for neurological functional recovery, protective effect of cell transplantation and possible mechanism.Methods:Get 150g SPrague an Dawley (SD) rat long bone marrow cells, through different training methods and then get MSCs and EPCs respectively ; using tied line method to make SD rat model(MCAO) (n = 100), and randomly divided into ischemic spontaneous recovery group (A group), PBS control group (B group), single transplanted MSCs group (C group) and transplantation MSCs and EPCs group (D group). SD rats with brain ischemia and reperfusion 24 hours after 2h, through the tail vein group D transplants 2×10~6 MSCs and 2×10~6 EPCs into MCAO rats' body; group C , transplanting graft with total volume equal to 2×10~6 into MCAO rats' body through the tail vein ; group B via the tail vein inject PBS equal to the total volume of fluid into the MCAO rats; Anaesthesia group A, without making any therapeutic treatment of skin suture after making isomeric model . 7 days later, observing neurological score, infarct volume, HE staining, brain-derived neurotrophic factor (BDNF) expression individually.Results:1. Cerebral Infarct Volume: Group A's infarct volume is (210.3±11.7) mm~3, Group B's infarct volume is (211.6±9.8)mm~3, Group C's infarct volume is(163.8±12.4)mm~3, Group D's infarct volume is(132.7±8.1)mm~3. Group D is distinctly differ from C, B, A about(P <0.05), and group C is distinctly differ from B, A about(P <0.05); There is no statistical significance between group B and A because P >0.05.2. Neurological Score: Group A with (3.78±0.49) marks, Group B with (3.78±0.50) marks, Group C with (3.78±0.49) marks, Group A with (3.78±0.53) marks. The comparison among these is obscure (P >0.05); The seventh days after transplanting, the rats' neurological functional recovery in the group D is (2.29±0.29), distinctly differ form group C (3.27±0.31), group B (3.74±0.31), group A(3.74±0.31) about (P <0.05), and group C is distinctly differ from B, A about(P <0.05); There is no statistical significance between group B and A because P >0.05.3. Pathomorphology(1) The BDNF in rat's brain of group A and B with less expression, more in the group C and D. Positive cytoplasmic's color is brown , while the nucleus was not stained, and more for the hippocampus and cortex of the pyramidal cells or granule cells. The BDNF expression in group D is more than group C. The results analysed by the SPSS system: In the seven days, the BDNF expression level of rat brain (22.0010±0.0948) is greatly higher than group C(19.1810±0.1128), group B(15.5441±0.0539), group A(15.5607±0.0481)about(P <0.05),and group C is distinctly differ from B, A about(P <0.05); There is no statistical significance between group B and A because P >0.05.(2)HE staining chips of brain tissue shows: a macrostructure microscope, group D has abundant cortical cells and cytoplasm, which cell damage is less than group B , and A. Through microscope, we can see a scattered small softening foci , and the relatively good remaining structure. Group B and A organizations have evident edema, and increasing volume of nerve cells, degeneration and cell gap; the group C is different from the other several groups because of poor organization, cell edema worse than the group D.Conclusions1: MSCs and EPCs transplantation treatment of ischemic brain injury may be superior to single transplantation MSCs, which can promote recovery of neurological function;2, MSCs and EPCs transplantation treatment of ischemic brain injury may be through promot the secretion of BDNF in brain, thereby improving nerve function.
