Objective:This study used bone marrow stromal cells bone marrow stromal cells, (BMSCs), endothelial progenitor cells (endothelial progenitor cells, EPCs) transplants Intervened the rat brain ischemia-reperfusion mode.To investigate the expressions of Bcl-2,Bax and the effect on bone marrow stromal cells(BMSCs) and endothelial progenitor cells(EPCs) transplantation for treating cerebral ischemia in rats.Methods:Rat middle cerebral artery occlusion(MCAO) model was established.A total of 100 SD rats were divided into sham operation group(A group),operation no treating group(B group),BMSCs(tail vein)group ,EPCSgroup and BMSCs +EPCScomplex group at random. A group of anesthesia, only isolated to the carotid artery, suturing the skin that is not any treated.B group is MCAO rat model.Ischemia reperfusion 2h, 7day after preparation of the model made, group training, respectively, BMSCs, EPCs dissolved in phosphate buffer solution, cell number was adjusted to 2×106 / L. C group is transplanted 2×106 / L BMSCs into the MCAO tail vein. D group is transplanted 2×106 / L EPCs into the MCAO tail vein.E group is transplanted 2×106 / L EPCs and2×106 / L BMSCs into the MCAO tail vein. Transplantation in each group equal to the total volume of liquid.After 7 days of reperfusion were observed neurologic score, HE staining, bcl-2 and bax.Results:1: Neurologic score: group A,B,C,D,E was 0,3.92±0.31,2.67±0.43,3.01±0.45分,1.92±0.25。repectively. Group B was significantly higher than group C,and group D showed a statistically significant difference(P<0.05), while group E showed a statistically significant difference compared with group C and group D.2:Immunohistochemical staining was used to detect the expression of Bcl-2: The expression level of Bcl-2 of group A,B,C,D,E is: 8.69±10.14,12.59±4.26,33.37±6.51,22.28±7.18, 45.67±5.02。group E was highest and showed a statistically significant difference(P<0.05)compared with the other groups, group C and group D showed a statistically significant difference compared with group B,and showed a statistically significant difference compared with group C and group D(P<0.05).3.Immunohistochemical staining was used to detect the expression of Bax:,The expression level of Bax of group A,B,C,D,E is: 7.15±8.50,39.89±8.46,21.32±3.91,28.57±4.67,10.49±7.24。.respectively, group B was the highest,group C and group D followed, group E was the lowest, and showed a statistically significant difference compared with group C and group D(P<0.05).4 . Brain histopathological slices stained with HE showed: with low power microscrope, group B organization edema, increased volume of nerve cells, vacuolar degeneration and cell space became larger, group E had more nerve cells, abundant cytoplasm, cell damage degree is the lowest of all groups. Small softening focus scattered,other organization structure is better.Conclusions:1: Bone marrow stromal cells(BMSCs)and endothelial progenitor cells(EPCs) Combined transplantation group,s therapeutic efficacy is more obvious than that's of BMSCs and and EPCs group. To better promote the repair of damaged nerve cells2: BMSCs and EPCs transplantation could improve the neurological function of brain injury rat, which may be related to up-regulating Bcl-2 and down-regulating Bax, and reducing apoptosis of neural cells.
