Effects Of Fresh Onion Extract On TNF-α Of Nonalcoholic Fatty Liver Disease In Clinical And Experiment Research

Objective:By observing effects of fresh scallion stalk extracts on NAFLD in patients with clinical efficacy, Biochemical index, B—ultrasonography and TNF-α, for the clinical application of Activating Yang Theory to provide a scientific basis.Methods:76 nonalcoholic fatty liver patients(including Tan ning xue yu from the First Hospital of Wuhan City) consistent with the diagnostic criteria were selected from 2008—12 to 2010—1, were classified according to the diagnostic criteria of Traditional Chinese Medicine. The patients randomized into treatment group(38 cases) and controlgroup (38 cases). The former was administered with Fresh Onion extract,2 capsules each, bid; the latter was given Essentiale,3 capsule, tid. Bothtreatment period were three months, two weeks after the with drawal to observe the effect. Before and after treatment were observed in patients with TCM syndrome, body mass index (BMI), blood lipids (TC, TG), liver function (ALT, AST,), B—ultrasonography points card and contents in serum TNF-α.Results:After statistical analysis, treatment group was 99.11% overall efficiency than the control group, the total efficiency of 73.68%; before and after treatment changes in body mass index and the B-were significantly improved, in each group after treatment of the TG, ALT, AST levels decreased, TNF-a levels significantly decreased after treatment.Conclusion:Fresh Onion extract can significantly improve symptoms in patients with fatty liver.Clear the liver cells, fat accumulation, recovery and improvement of liver imaging. Regulate lipid metabolism to reduce body mass index, improve liver function and blood lipids, reduce inflammation in liver tissue. Effective treatment for NAFLD, its mechanism may be related to inhibiting TNF-a expression, reduce its activity.Objective:Observation of fresh scallion stalk extracts on TNF a with fatty degeneration of the liver cell model, and to explore the mechanism of its prevention and treatment NAFLD. Methods:With free fatty acid-induced HepG-2 cell model for the establishment of liver cell steatosis. Grouped under the normal group, model group, different concentrations of drug groups. Using oil red O staining of lipid droplets within cells, and to detect intracellular TG, ELISA to detect TNF-αlevels.Results:After 24h free fatty acid-induced, Oil red O staining showed that fatty degeneration of HepG-2 cell, TG content in model group were significantly higher than the control group(P<0.01), the supernatant levels of TNF-αincreased significantly(P<0.01).The extract of fresh scallion stalk the role of different doses of drugs within the group of intracellular lipid deposition, TG was significantly lower than that of control, and to reduce the level of TNF-α, compared with model group differences were statistically significant(P<0.05).Conclusion:Fresh Onion extract on fatty degeneration of the liver cells has obvious protective effect, the mechanism may be related to decreased expression of TNF-α.In summary, this study shows that fresh light blue Tong Yang Chinese medicine extract can suppress TNF-αexpression, and thus anti-fatty degeneration of liver cells and improve the role of cellular energy metabolism. Thus reveal the fresh scallion stalk extract effective treatment for NAFLD, its mechanism may be related to inhibiting TNF-αexpression, reduce its activity.
