Objective:To investigate whether onion alcohol extract(ACE)has a lipid-lowering effect on rat models of hyperlipidemia,and its mechanism and active ingredients.Methods:Sixty male SD rats were randomly divided into six groups:Normal group(NC),high-fat group(HF),Positive group(PC),low-dose group(AC-L),medium-dose group(AC-M),high-dose group(AC-H),There are 10 SD rats in each group.The NC was fed with basic feed and saline,and the others were fed with high-fat diet(HFD),high-fat emulsion(HFE).Two weeks later,different dose of ACE intervention experiments were conducted,and two weeks after drug intervention,the rat samples were taken.(1)Corresponding kits detect total cholesterol(TC),triglyceride(TG),low-density lipoprotein(LDL),and high-density(HDL),superoxide dismutase(SOD),and malondialdehyde(MDA)changes in SD rats;calculate the Atherosclerosis Index(AI)and assess the risk of atherosclerosis;(2)HE staining to observe morphological changes in aortic arch and liver;(3)Western-blot test HMGCR and LDLR protein expression;(4)HPLC was used to detect the contents of quercetin and isoquercetin in ACE.Results:(1)Compared with HF,ACE can reduce the levels of TC,LDL and MDA(P<0.05),increase the levels of HDL and SOD(P<0.05),in the serum of hyperlipidemia rats,and reduce the value of AI(P<0.05)),the change of TG is not obvious(P>0.05);Decrease the levels of TC,TG,LDL,and MDA(P<0.05)and increase SOD levels(P<0.05),in liver tissues of hyperlipidemia rats;(2)ACE can make liver tissues caused by hyperlipidemia The degree of aortic arch disease was reduced;(3)Compared with HF,the relative expression of LDLR in liver tissue in AC-L,AC-M,and AC-H increased by 49.1%,72.7%,and 100.0%and the relative expression of HMGCR protein Decrease by 21.1%,30.0%,and 45.5%,respectively.(4)ACE contains quercetin and isoquercetin,and their contents are 2.42 mg/g and 4.60 mg/g,respectively.Conclusion:(1)ACE can reduce blood lipid levels in the hyperlipidemia rat model and has certain protective effects on the liver and aortic arch;(2)ACE exerts its lipid-lowering effect by increasing the expression of LDLR protein and reducing the expression of HMGCR protein;(3)Active ingredients that exert lipid-lowering effects in ACE may include quercetin and isoquercetin.
