Objective To ascertain the effect of change traction on articular cartilage by utilizing llizarov distraction technique of minimally invasive.Methods Experiment at 18 dogs (randomly divided into two groups-traditional Chinese medicine Fumigation group, external fixation group) left ankle joints were fitted with Ilizarov external fixator, the use of lmm/d of the stretch speed, firstly create a varus deformity, three weeks time, to the ankle varus 15°,taken X-ray film confirmed after the cessation of stretch three weeks, and then the same speed (lmm/d), to correct deformities, three weeks time. And in the first 3 weeks,7 weeks,9 weeks were collected the experimental medial articular cartilage and articular cartilage health medial. To observe the changes in the microstructure of the articular cartilage by using Transmission electron microscopy and Multi-photon laser scanning confocal microscope.Results 1.Observed by transmission electron microscopy.The first 3 weeks of cartilage cells become flat.There are mainly euchromatin. Some of chromatin appeare under the nuclear membrane.In the cartilage cells.The nomber of organelles decreases.Golgi complex, rough endoplasmic reticulum, mitochondria decrease, but the structure is still complete,but the structure is still complete. There are a small amount of lipid droplets.Collagen fiber bending becomes thin, loose change, but the arrangement of light and dark stripes and white still rules; The 7 weeks of cartilage cells become more deformation. In the cells,nuclear is obviousely deviating and withering. Most of uptake of chromatin appeare under the nuclear membrane.The nomber of organelles significantly decreases. The structure of Golgi complex, rough endoplasmic reticulum, mitochondria is damaged. Golgi complex relaxation expansion andthere are degranulation.Most number of mitochondria disappear,double-film of mitochondria vague and a number of nuclear pores Appear. Lysosome lysosome and second terminal can be seen.The fracture of Collagen fiber bending becomes more loose and disordered; The 9 weeks cartilage cells are mostly flat and protruding more nuclear deviation.Margination of chromatin apper under the nuclear membrane. The number of organelles is reduction.Golgi complex, rough endoplasmic reticulum, mitochondria and other organelles decreases and the structure of Golgi complex, rough endoplasmic reticulum are clear and complete.Double-membrane structure of mitochondrial is complete and Degranulation disappear.Arrangement of collagen fibers appeare certain texture, more porous, smaller.2. Observed by multi-photon laser scanning confocal microscope:The 3 weeks cartilage cells fluorescent red and the color fades.The number of vision reduce. Some structure of cartilage cells are cracking. Collagen fibers fluorescent pale green and the color pale, yet orderly arrangement; The 7 weeks cartilage cells fluorescent dark gray.The number of vision markedly reduces.Most of the structural are damaged. Collagen fibers fluorescent green and arrange in disorder; The 9 weeks cartilage cells fluorescent red. The number of cartilage cells decrease.Some cells split regeneration. Collagen fibers is dark green, arranged in a certain coherent beginning.Conclusions:Traction at ilizarov stretch technology, when the pressure between the cartilage surface of the ankle joint increases, the articular cartilage degeneration occurs and when the stress between the articular cartilage decreases.the articular cartilage regeneration can be able to restore.
