Objective:Colorectal cancer(CRC) is one of the common malignant gastrointestinal cancers in China. Currently the disease incidence is rising year by year. However,the mechanisms of CRC development remain to be studied.This study aimed to detect the inflammatory markers, including MMP-9, COX-2, and KLF5, which plays an important role in inflammation and tumors,and to explore the correlation between the inflammatory markers and progression of colorectal cancer.Methods:1. PCR was used to detect MMP-9 gene R279Q genotypes.2.Zymography was used to examine the activity of MMP-9 in the cancer tissues and the matched normal tissues.3.1mmunohistochemical staining was used to detect the expressions of MMP-9, COX-2and KLF5 in colorectal carcinoma.Results:1 The polymorphism of MMP-9 gene Q279R ang -1562(C/T) were not associated with the development of CRC.There were three genotypes of MMP-9 Q279R including AA,AG and GG in patients with CRC and controls.The frequency of the A allele was not significantly different between CRC patients and the controls.(P=0.896).In addition,PCR-SSP showed that polymorphisms of MMP-9 gene-1562 (C/T) had CC,CT and TT genotypes,which was not correlated with developm-ent of CRC. The frequency of the T allele was not significantly different between CRC patients and the controls.(P=0.538).2. The activity of MMP-9 is increased in CRC tissuesThe result of zymography showed that the activity of MMP-9 in the can- cer tissues was significantly higher than that in the matched normal tissues (P<0.05).The expression of MMP-9 was significantly higher in Dukes C stage than those in Dukes A stages (P<0.05)3. COX-2 expression was positively correlated with the Dukes stagesImmunohistochemistry results showed that the expression of COX-2 was significantly higher in Dukes C stage than those in Dukes A stages (P<0.05)4. KLF5 expression was significantly correlated with the Dukes stagesImmunohistochemistry results showed that the expression of KLF5 was significantly higher in Dukes C stage than those in Dukes A stages (P<0.05)Conclusion:1 Polymorphisms of MMP-9 gene Q279R and -1562(C/T) are not associated with the risk of CRC.2 There is positively correlation between the expression of inflammatory markers,including MMP-9, COX-2 and KLF5, and the development of CRC.
