Objective: Psoriasis (PS),also known as"psoriasis"is a common and easy to relapse of chronic inflammatory skin disease. Due to a higher incidence of this disease,easy to relapse,a longer course of disease, particularly in young adults for multiple violations,so the patient's physical health and mental impact of very large. Epidermal hyperplasia and dermal papilla layer of angiogenesis is the main pathological features of psoriasis. Genetic factors and environmental stimuli are the two main factors of its onset.The complexity of clinical manifestations of psoriasis for many genetic diseases, its incidence related to the following four major cell types: keratinocytes, lymphocytes, endothelial cells and neutrophils, in which the two most crucial before. Most scholars believe that psoriasis is a T cell-mediated chronic inflammatory diseases. Psoriatic lesions, the epidermis and dermis has confirmed the existence of activated T lymphocytes.Inhibit T-cell activation directly inhibit the formation of psoriatic skin lesions of psoriasis and normal skin.Psoriasis of activated T cells can release inflammatory factors,such as TNF-a,IL-8 and INF-and so on, the promotion of psoriatic keratinocytes in cell proliferation and neutrophil migration;same time,T cell activation and neutrophil chemotaxis and the formation of a T cell-mediated inflammatory response positive feedback Loop.CD147 molecule is an immunoglobulin superfamily belonging to the transmembrane glycoprotein,is a high in vivo keratinocyte proliferation and poorly differentiated state marker, has been found that CD147 and T cell development,differentiation,especially activation of close related.New research shows that, CD147 may be by stimulating the secretion of hyaluronic acid-induced activation of CD4 10 T cells,CD147 can promote monocyte-macrophage cell maturation and cytokine secretion,to enhance inflammatory tissue monocytes/macrophages infiltration.CD44 is a cell surface integral membrane protein,is a family of adhesion molecules, it has a wide range of biological functions.CD44S and other molecular studies have shown that lymphocyte activation in the provision of costimulatory signals, in particular,such as T lymphocytes and NK activation. During the activation process, CD44 play a"co-stimulatory molecule"role of CD44 molecules on the membrane and its ligands hyaluronic acid or the corresponding antibody (anti-CD44 monoclonal antibody) combined,as a co-stimulatory molecule with the TCR/CD3 complex stimulation of acceptable together cause the proliferation of peripheral blood T cells to Th cells, the release of IL-2, CTL-induced release of pancreatic protein esterase.In order to understand psoriasis CD147,CD44 expression and the determination of hyaluronic acid in the blood, we used immunohistochemistry to detect the CD147,CD44 in psoriasis patients and lesions in different phases of expression, and RIA Determination of hyaluronic acid content of blood and thus to explore their role in psoriasis.Methods: Specimens from skin lesions in October 2006-2009 year in October the Fourth Hospital of Hebei Medical University,dermatology out-patient clinic patients, based on clinical performance,newly diagnosed patients with psoriasis in skin lesions under local anesthesia with the Department of trephine drawn,immediately placed in 10% formaldehyde solution, embedded in paraffin and reserve.The combination of selected clinical pathological specimens confirmed 32 cases of psoriasis patients,18 cases were male and 14 female; aged 10-80 years, mean 27.2 years of age.To exclude all patients suffering from other systemic diseases,and nearly 3 months had not received any treatment. Specimens from the control group,the Fourth Hospital of Hebei Medical University,20 cases of traumatic surgery in patients undergoing the health of the skin,control patients did not suffer from other diseases,and age,sex,and drawn parts to match patients with psoriasis. Histopathology using HE staining of paraffin sections,immunohistochemistry with SP method,staining positive staining for the brown granules. Application SPSS13.0 statistical software analysis test results. Immunohistochemical staining using rank sum test Mann-Whitney U tests and analysis of the indicators in the PS group and the control group,expression, CD147 and CD44 correlation with Spearman rank correlation analysis.Results:1 CD147 in normal skin keratinocytes and in psoriasis groups (PP, PvP, and PvS group) lesions keratinocytes,T cells and neutrophils were expressed. In 32 cases of psoriasis,the positive expression of a total of 26 cases,of which 8 cases were strongly positive,positive,weakly positive for all 9 cases and negative in 6 cases.The normal control group, negative in 18 cases,weakly positive in 1 cases,positive in 1 case,no strong positive expression.The results showed that,CD147 expression in psoriasis than those in the normal control group increased,the difference was significant (P<0.05),and advanced in the expression of psoriasis is higher than resting,the difference was significant (P <0.05).in different sex,age and course of disease there was no significant difference between (P>0.05).2 CD44S in normal skin and psoriasis lesions appearance of the Agency and its adjacent normal skin were expressed, including cells in the epidermal cells of the spine and the basal cell surface expression of granulosa cells and cuticle cells were No expression of cell surface expression in the spine strongest. While the control group only 1 case in 20 patients with positive expression, 2 cases of weak positive expression, the remainder being negative. The results showed that, CD44 expression in psoriasis than those in the normal control group increased, the difference was significant (P <0.05), and advanced in the expression of psoriasis is higher than resting, the difference was significant (P < 0.05). in different sex, age and course of disease there was no significant difference between (P> 0.05). 3 We clearly expressed the epidermis CD147-positive specimens of 26 patients with positive group, and another group of 6 cases of a clear expression of CD147-negative specimens of the control group. CD147 positive group was higher than that of hyaluronic acid.4 CD147 and CD44 in psoriatic lesions in patients with the same expression of the correlation coefficient r = 0.474, the difference was significant, positive correlation between the two.Conclusion: According to the experimental results the following conclusions:1 CD147 was mainly expressed in the psoriatic keratinocytes, T cells and neutrophils and was significantly higher, and advanced lesions than quiescent. Tip psoriatic lesions and increased levels of CD147 expression in disease classification and progress of the degree of correlation.2 CD44S expression in psoriasis vulgaris mainly located in epidermal cells and basal cells of the spine, in the spine on the cell surface expression of the strongest; dermal tissue showed uniform expression of CD44S; in the skin appendages in the hair follicles, sweat glands and blood vessels of small were expressed in endothelial cells. And significantly higher, and advanced lesions than quiescent. Tip CD44S psoriatic lesions and the expression level increased with the progress of the disease sub-type and degree of correlation.3 CD147 and CD44 positive correlation, suggesting that the role of hyaluronic acid in psoriasis, CD147 secretion of hyaluronic acid, and its receptor CD44S combined as co-stimulatory molecule with the TCR/CD3 complex with the accepted cause stimulation of peripheral blood T cell proliferation.4 psoriasis CD147-positive group of hyaluronic acid was significantly higher than that CD147-negative, CD147 positive correlation with the hyaluronic acid, suggesting that CD147 and hyaluronic acid generate information.5 CD147, CD44S gender, age, and no correlation between duration.
