Objective:1 Observing the morphologic variety of the placental villus durning 15days before pregnancy and early trimester of pregnancy exposure to the BaP;2 Studying the mechanism of action of damnification that BaP act on placental villus;3 Studying the effect of Vitamin E oppose BaP;4 Providing the guidelines of antenatal care and clinical research.Methods:1 Choosing of the experimental animals:150 ICR mice(100 female mice,50 male mice) were randomly divided into five groups, each with30(20 female mice,10 male mice),and raised female mice and male mice respectively.2 Grouping the experimental animals The control group: female mice was lavaged with BaP(2mg/kg.d); 10mg/kgVE ant-group:female mice was lavaged with BaP(2mg/kg.d)and Vitamin E(10mg/kg.d); 20mg/kgVE ant-group:female mice was lavaged with BaP(2mg/kg.d)and Vitamin E(20mg/kg.d); 40mg/kgVE ant-group:female mice was lavaged with BaP(2mg/kg.d)and Vitamin E(40mg/kg.d); The vacuity group:female mice was lavaged with olive oil in identical capacity everyday.3 Dealling with the experimental animalsAfter 15days(three estrous cycles),male mice were mate with female mice(every cage included 2 female mice and 1 male mouse),The second day,Gestational age was determined based on the presence of a vaginal plug, with the morning of detection being embryonic day 1 (d 1) of gestation,we did no treatment on male mice. Electing pregnant mice in each group. we continued lavaging BaP(2mg/kg.d)for 7 days(early trimester of pregnancy include 7 days).The mice were euthanized then detected the relevant indicators.4 Method of detecing related index4.1 Detecting the gene of Bax and Bcl-2 in the level of mRNA with RT-PCR in the placental villus;4.2 Detecting activity of SOD and GSH-PX with related kit;4.3 Observting the morphology change with HE;4.4 Detecting the gene of Bax and Bcl-2 in the level of protein and observting the location of the protein of Bax and Bcl-2 with .4.5 Statistical analysis using software of SPSS10.0Result:1 The expression of Bax and Bcl-2 relate to apotosis of placental villus in the level of mRNA: Array lavaged with BaP(2mg/kg.d)expressed more Bax in the level of mRNA than lavaged with olive oil(P<0.01).Lavaged with different dose of VE(10mg/kg.d 20mg/kg.d 40mg/kg.d),the expression of Bax in the level of mRNA become less,but 10mg/kg.d and 20mg/kg.d have not statistics significant(P=0.183>0.05), 10mg/kg.d and lavaged with BaP have not statistics significant(P = 0.197 > 0.05). Array lavaged with BaP(2mg/kg.d)expressed less Bcl-2 in the level of mRNA than lavaged with olive oil(P<0.01), Lavaged with different dose of VE(10mg/kg.d,20mg/kg.d, 40mg/kg.d),the expression of Bcl-2 in the level of mRNA become more(P<0.05),the more VE,the more Bcl-2 in the level of mRNA(P<0.01). The more VE,the less Bax/Bcl-2 in the level of mRNA(P<0.05).Bax and Bcl-2 have relationship(r=-0.951,P=0.002<0.01).2 The activity of GSH-PX and SOD in the placental villus:The activity of GSH-PX and SOD of array lavaged with BaP(2mg/kg.d) less than array lavaged with olive oil(P<0.01). Lavaged with different dose of VE(10mg/kg.d 20mg/kg.d 40mg/kg.d),the activity of SOD and GSH-PX become more(P<0.05).The more VE,the more activity of SOD and GSH-PX in the level of mRNA(P<0.05).3 Observing the morphological variety of the placental villus with HE:BaP made the cells of placental villus apotosis,our experiment observed the morphological variety of the placental villus in 400×microscope with HE.The cells of placental villus represented apotosis characteristic including:karyotheca crimple,karyon nucleolus crimple,karyon nucleolus cataclasm. Lavaged with different dose of VE(10mg/kg.d 20mg/kg.d 40mg/kg.d),the number of apotosis cells decreased,The more VE,the less number of apotosis cells.4 The expression of Bax and Bcl-2 relate to apotosis of placental villus in the level of protein: Array lavaged with BaP(2mg/kg.d)expressed more Bax in the level of protein than lavaged with olive oil(P<0.01),expressed less Bcl-2 in the level of protein than lavaged with olive oil(P<0.01). Lavaged with different dose of VE(10mg/kg.d 20mg/kg.d 40mg/kg.d),the expression of Bax in the level of protein become less(P<0.05) ,the expression of Bcl-2 in the level of protein become more(P<0.05). The more VE,the less Bax/Bcl-2 in the level of protein(P < 0.01).Bax and Bcl-2 have relationship(r=-0.825,P=0.021<0.05).The expression of Bax gene in the level of mRNA and protein have relationship(r=0.789,P=0.025 < 0.05), the expression of Bcl-2 gene in the level of mRNA and protein have relationship(r=0.972,P=0.018<0.05).Conclusion:1 The mice expose to BaP(2mg/kg.d) can make the the cells in the placental villu apotosis durning the period from Progestation(15 days) to early trimester of pregnancy(7 days);2 Our expremment futher confirmed that BaP can make the the cells in the placental villu apotosis through the path of increasing the expression of Bax in the level of mRNA and protein,decreasing the expression of Bcl-2 in the level of mRNA and protein; 3 Our expremment futher confirmed that BaP can make the the cells in the placental villu apotosis through the path of decreasing the activity of SOD and GSH-PX;4 VE can suppressed the the cells in the placental villu apotosis through the paths of decreasing the expression of Bax in the level of mRNA and protein, increasing the expression of Bcl-2 in the level of mRNA and protein,and increasing the activity of SOD and GSH-PX.
