| Objective:1. To test the expressions of STAT3,STAT3 mRNA in human gastric carcinoma and normal gastric mucous;2. To evaluate the regulationship that EGFR,SOCS1 toward STAT3;3. To analyse the expression of Survivin,Bcl-2,VEGF in gastric carcinoma and nomal gastric mucous,and evaluate the correation between these proteins and STAT3;4. To analyse the relationship between above mentioned proteins and various clinicopathology factors, prognosis.Methods:The clinicopathologic data of 53 gastric carcinoma patients were analyzed retrospectively. All patients underwent radical gastrectomy in Tianjin Medical University in January 2001 to December 2001. We also obtained 53 nomal gastric mucous as contrast. The expression of STAT3 mRNA were detected by in situ hydration. The expression of STAT3,EGFR,SOCS1,Survivin,Bcl-2,VEGF were determined by immunohistochemical staining of tissue from gasric cancer specimens and normal gastric mucous. We evaluated the relationship between expression of above mentioned proteins and various clinicopathological factors.Results:1. The positive expression rate of STAT3,STAT3mRNA in the gastric cancer specimens were significantly higher than those in the normal gastric mucous: (58.5%VS11.3%,62.3%VS9.4%) (P<0.001). There were a positive correlation between the positive expression rate of STAT3 and STAT3mRNA in the human gastric cancer tissues (r=0.766, P<0.001);2. There were a significantly positive correlation between the positive expression rate of STAT3 and EGFR in the human gastric cancer tissues,r=0.371(P=0.007), whereas there were a negative correlation between the positive expression rate of STAT3 and SOCS1 in the human gastric cancer tissues, r=-0.424 (P=0.002);3. The positive expression rates of Survivin,Bcl-2,VEGF in 53 gastric carcinoma tissues were 73.6%(39/53),49.1%(26/53),62.3%(33/53), respectively. They were significantly higher than those in the normal group: 7.55%(4/53),0%(0/53),24.5%(13/53)(P<0.05). STAT3 expression correlated with Survivin(r=0.451), Bcl-2(r=0.597), VEGF (r=0.608)(P<0.01);4. Positive stainings of STAT3 and VEGF were significantly associated with tumor size, Lauren's classification, lymph node metastasis and clinical staging(P< 0.05), Positive stainings of Survivin was significantly associated with Lauren's classification, lymph node metastasis and clinical staging(P<0.05). Positive stainings of Bcl-2 staining was significantly associated with tumor size, lymph node metastasis and clinical staging(P<0.05);5. With univariate analysis, the preditors of survival in gastric cancer patients were STAT3 expression,Survivin expression,Bcl-2 expression,VEGF expression,tumor size, lymph node metastasis and clinical staging. With multivariate analysis, STAT3 expression and lymph node metastasis were independently prognostic factors of poor survival.Conclusion:1. STAT3 mRNA and STAT3 were over-expressed in human gastric carcinoma,which play the important roles in the carcinogenesis and progression of gastric carcinoma;2. The EGFR could enhance STAT3 through the JAK and/or non-JAK pathway. The effect of SOCS1 in gastric cancer tissues is negative feedback toward JAK-STAT signal pathway, it's the reason of continuous activation of STAT3ï¼›3. The activation of STAT3 could improve the expression genes of cell apoptosis(Survivin,Bcl-2), and the gene of angiogenesis(VEGF).The over-expressed STAT3 and related proteins are associated with poor biological behaviour;4. The positive expression of STAT3 was independently prognostic factor in gastric carcinoma. |
