| Objective:To investigate the changes of endothelin-1 (ET-1) in serum and the regulation in acute myocardial ischemia-reperfusion injury model and the protecting effects of L-arginine.Methods:100 adult healthy wistar rats were randomizily divided into control group (Group A), ischemic 30minutes group (Group B), ischemic 30minutes+ reperfusion 0.5 hour group,ischemic 30minutes+reperfusion 1 hour group(Group C), and L-arg+ischemic 30minutes,L-arg+ischemic 30minutes+reperfusion 1 hour group (Group D), L-arg+ischemic 30minutes+reperfusion 2 hour group, L-arg+control group. The activity of CK and LDH were determinated by Enzyme dynamic method.The changes of ET-1 were determinated by ELISA method and Western blot methods. To measure the activity of CK and LDH with Dynamic Determination of enzyme.Results:The improved method can has a good success rate versus conventional method. MI/RI causes the dysfunction of endothelium. The serum levels of ET-1 is increased significantly in acute myocardial ischemia-reperfusion injury and the levels of ET-1 in group C is higher than that in group B, group B is higer than that in group A.In RT-PCR compared with theβ-actin the ratio of the control group (Group A), ischemic 30minutes group (Group B), ischemic 30minutes+reperfusion 0.5 hour group,ischemic 30minutes+reperfusion 1 hour group (Group C), ischemic 30minutes+reperfusion 2 hour group, and L-arg+ischemic 30minutes+reperfusion 1 hour group,L-arg+ischemic 30minutes+reperfusion 2 hour group (Group D), L-arg+ ischemic 30minutes and L-arg+control group is 0.31,0.39,0.52,0.73,0.775,0.505,0.55,0.34,0.556; compared with theβ-actin the ratio of the A, B,C and D group is 0.52,0.67,0.94,0.58. The lever of ET-lin Group B, Group C were increased significantly (P<0.05),but in Group A, and Group D there were no significant difference (P>0.05). L-arg can reverse the damage of ET-1.In western blot compared with the GAPDH the ratio of thecontrol group (Group A), ischemic 30minutes group (Group B), ischemic 30minutes+reperfusion 0.5 hour group,ischemic 30minutes+reperfusion 1 hour group (Group C), ischemic 30minutes+reperfusion 2 hour group, and L-arg+ischemic 30minutes+reperfusion 1 hour group,L-arg+ischemic 30minutes+reperfusion 2 hour group (Group D), L-arg+ ischemic 30minutes and L-arg+control group is 0.31,0.39,0.52,0.73,0.775,0.505,0.55,0.34,0.556; compared with the GAPDH the ratio of the A,B,C and D group is 0.52,0.67,0.94,0.58. L-arg can reverse the damage of ET-1.Conclusion:1.The serum levels of endothelin-1 (ET-1) were increased in acute myocardial ischemia-reperfusion injury model and the levels of ET-llevelswere increased significantly in Group B and Group C.2.The expression of Endothelin-1 is an important indicator to diagnosis the injury of myocardial ischemiac-reperfusion injury.3.L-arg can reversr the change of ET-1and protect the coronary endothelium. 4. As a protecting measures of myocardial ischemia reperfusion injury,L-arg was verified in the model of rats.In couclusion,ET-1 was increased in myocardial ischemia and reperfusion injury in rats. L-arg could reduce ET-1 in levels of, mRNA and protein at certain stages. |
