Clinical Exploration On Hematologic Malignancies Treated By Autologous Peripheral Blood Stem Cells

Objective:To observe the activity of autologous peripheral blood stem cells (PBSC) cryopreserved directly at-80℃and to observe patients' hematopoietic reconsti-tution after hematologic malignancies (malignant lymphoma, acute leukemia) treated by autologous peripheral blood stem cell transplantation (APBSCT). Methods:Patients' autologous PBSC were collected by CS3000 Plus blood cell separator for 1 to 3 times to achieve the required number of transplantation. Then the PBSC were cryopreserved directly at-80℃refrigerator. Before cryopreservation and after reinfusion, we collected samples for detecting mononuclear cells (MNC) counts,the number of CD34+cells and trypan blue exclusion. And we monitored the change of peripheral blood phenomenon after transplantation. Results:Before cryopreservation, the averages of MNC,CD34+ cells and trypan blue exclusion were (4.91±3.03)×108/kg,(3.32±2.45)×106/kg,(95.20±1.68)%, respectively; after reinfusion, the averages MNC,CD34+cells and trypan blue exclusion were (4.16±2.55)×108/kg,(2.86±1.98)×106/kg and (83.50±2.32)%, respectively. There were statistical differences between the results after reinfusion and the results before cryopreservation to MNC counts,the number of CD34+cells and trypan blue exclusion (p<0.05).The recovery rates of MNC and CD34+cells were 84.73%,86.14%, respectively. All the patients had gotten hematopoietic reconstructtion after transpl-antation. And Leukocyte recovery (>1.0×109/L) time was (10.80±1.25) days,neutrophils recovery (>0.5×109/L) time was (10.40±1.13) days and platelet recovery (>20×109/L) time was (13.30±4.09) days. Conclusions:APBSCT was a convenient and effective method to used to the treatment of hematologic malignancies. The recovery rate of PBSC cryopreserved directly at-80℃can meet the needs of transplantation. And all patients can be got hematopoietic reconstitution after transplantation. But the interference effects of some factors affecting cryopreservation require further attentiom.
