Objective:Rats experimental periodontitis, type 2 diabetes, type 2 diabetes periodo-ntitis model was established respectively. The expression level of OPG and RANKL was detected in ingingival crevicular fluid(GCF) and blood serum each group. The two cytokines was detected in GCF after periodontal initial therapy together with local injection of recombinant human osteoprotegerin (rhOPG) into the periodontal tissue of rats experimental periodontitis. The aim of the research is to clear wheth-er OPG and RANKL may play a role in periodontitis and type 2 diabetes pathoge-nesis or not, so as to provide the foundation of appling rhOPG in the treatment of periodontitis.Methods:Experimental rats were randomly divided into four groups, P group Rats were given wire around the cervix of the second maxillary molars and provided w-ith drinking water containing 10% glucose. D group Rats were fed with high sucr-ose-fat diet and low-dose STZ was injected into abdomen after 6 weeks to establih rat model of type 2 diabetes mellitus. DP group rats were disposaled with P+D method to build the experiental type 2 diabetic periodontitis model. N group was control group. After establishing all the models successfully, ELISA was used to detect the expression of OPG and RANKL in GCF and blood serum. Taking 10 r-ats from P group randomly, Remving the ligature wire around the cervix of the se-cond maxillary molars and performing subgingival curettage for the rats, the rats were randomly divided into 2 groups, a group rats were injected rhOPG into perio-dontal tissue,2 times a week, while b group rats were no-disposalling. Then ELIS-A was used to detect the expression of OPG and RANKL in GCF.Results:1.The expression of OPG in GCF of control group was increased obviously than those in simple periodontitis group and type 2 diabetic periodontitis group and the expression of RANKL was decreased obviously in GCF of control group than those in the simple periodontitis group and the type 2 diabetic periodontitis group (P<0.05).2. The expression of OPG and RANKL in blood serum of type 2 diabetes group and type 2 diabetic periodontitis group were increased obviously than those in the simple periodontitis group and the control group (P<0.01).3.Comparied to pre-and after-periodontal therapy, the expression of OPG in GCF was significant increased after periodontal initial therapy, and the expression of OPG in the group with periodontal initial therapy together with local injection of rhOPG was increased obviously than the group with periodontal initial therapy (P<0.05). However, the expression-of RANKL in GCF is just the opposite.Conclusion:1. The expression of OPG was decreased, while the expression of RANKL was increased in GCF of type 2 diabetic and periodontitis. It is suggested that the imbanlance of OPG and RANKL may be main reason resulting in periodontitis, Type 2 diabetes as the general factor may cause the host high susceptibility to periodontitis and accelerate the imbalance of OPG and RANKL, Therefore, it may promote the occurrence and development of periodontitis.2. The expression of OPG and RANKL were increased in blood serum of type 2 diabetes. It is suggested that type 2 diabetes may influence the expression of OPG and RANKL in GCF by changing the hormones and cytokines of blood but not affecting periodontium directly.The expression of OPG and RANKL in GCF is influenced not only by inflammation and infection but also blood sugar.3. The chang of OPG and RANKL was significant between the group of periodontal initial therapy together with local injection of rhOPG and the group of periodontal initial therapy, It suggests that the studies will provide the experimental basis for understanding the reason of periodontitis with type 2 diabetes and improving the therapy of periodontitis.
