Objective:To investigate the feasibility of cervical mucus exfoliated trophoblast cells in prenatal disgnosis, by analyzing the qualitative of fetal cells of early pregnancy in cervical mucus with method of Real-time PCR. Methods:160 samples of cervical mucus were obtained in the gestational age of 6-10 weeks just before the artifical abortion, further more,the fetal DNA were extracted from the mucus. Then sex Y chromosome special sequences DYS14 as the marker of fetal DNA were amplified by real-time PCR,β-globin gene as an internal control. The sex and chromosomal karyotype of fetus were determined by chromosome karyotyping of the villi. Results:The six Y chromosome sequences were detected in 68 of 76 male carrier samples, and 11 of 84 female carrier samples. The sensitivity was 89.5% and the specificity was 86.9% in determination of fetal gender during the first trimester. Conclusion:Transcervical cells sampling represents a promising prospect for non-invasive prenatal diagnosis. The accuracy and reliability are yet to be improved.
