Objective:To establish dogs model of severe myocardial contusion in opened and prelimi-nary study on its metabolism change of glucose in heart after SMC, FDP were observed in dogs without treatment after SMC. Methods:1. Sixteen healthy mongrel dogs were randomly divid-ed into four groups, groupⅠ(n=4,control group) received impact with Okg weight, groupⅡ(n=4) received impact with 1.0kg and diameter 5cm weight fall of at 85cm, groupⅢ(n=4) received impa-ct with 1.25kg and diameter 5cm weight fall of at 85cm, and groupⅣ(n=4) received impact with 1.5kg and diameter 5cm weight fall of at 85cm. The dogs were injured with weight falls down by free fall movement to the surface heart, observed blood press decrease level after impact with per dog. Blood coronary vein was coollected at pre-impact 30mins, post-impact at 30mins, 1h,2h,4h and 6h, checked thickness of MDA, CK-MB and cTnI. The myocardium were coollected from the impact part area 0.8cm×0.8cm were taken for histopathological examination after executded.2. weight healthy mongrel dogs were randomly divided into contusion groups (n=4) and treatment group(n=4), impact dogs heart with the mass, diameter and height maked into SMC in opened, keeped circulate steady in contusion group by infusion NaCl and beside infusion FDP with 1300 mg/kg in treatment group at 30min after impact. Blood of arterial and coronary vein were coollect-ed at pre-impact 30mins, post-impact at 30mins, 1h,2h,4h and 6h, checked thickness of Glu, FFA,GH and TNF-a.Monitoed of LVSP, LVEDP and±dp/dtmax at same time point. Results: (1). Set up the model of dogs SMC. MAP undecrease after SMC of dogs in groupⅠ. plasma of CK-MB, cTnI and MDA were increase lightly. histopathological result showed:myocardial was normal.Compared with pre-impact, MAP decrease after SMC of dogs in groupⅡ,none of them downed (>30%), plasma of CK-MB, cTnI and MDA were increase lightly. Histopathological result showed:there were haemorrhagia of myocardial, undisorder of the myocardial fibers, some hemorrhagic spot.Compared with pre-impact, one of them MAP decrease(>30%) after SMC of four dogs in groupⅢ, plasma of CK-MB, cTnI and MDA were increase apparently(P<0.05).histo-pathological result showed:There were haemorrhagia of myocardial, congestion, swelling,and undisorder of the myocardial fibers, even some quantity neutrophil and lymphocyte were infiltrat-ed. Compared with pre-impact, MAP decrease apparently after SMC(>30%) of four dogs in groupIV, plasma of CK-MB, cTnI and MDA were increase apparently (P<0.01).histopathological result showed:There were widely-spreaded haemorrhagia of myocardial, congestion, swelling and broken of the myocardial fibers, even a great quantity neutrophil and lymphocyte were infilt-rated. (2) Myocardial glusose metabolism:Plasma glucose in two groups increase apparently after SMC, and all decrease hereafter, and remaining at high level until 6 hours post-impact (P<0.05). Net myocardial glucose ingestion and myocardial ingestion ratio in MC group descend to zero at 1 hours after impact. Compared with MC group, myocardium glucose uptake/utilization are exis-tence at all time in Therapy group, and decrease lowest at 1 hours after impact (P>0.05), increase hereafter, there were obvious discrepancy (P<0.01) at 6 hours after impact. (3) Change of Free fatty acids:Free fatty acids are increased obviously in two group after SMC. Compared with MC group, which increase more obviously until 6 hours after impact in Therapy group(P<0.01). (4) Growth hormone, Tumor Necrosis Factor-α(TNF-α):Plasma growth hormone and TNF-αin two groups increase apparently after SMC, and all decrease hereafter, and remained at high level until 6 hours after impact(P<0.05). Compared with MC group, which decrease more apparently in Therapy group, there were obvious discrepancy(P<0.05) at 6 hours after impact.(5) Hemodynami-cs:Left ventricular systolic pressure (LVSP) and maximal rate of rise of left ventricular pressure (LV dp/dtmax) in two groups all decrease after SMC, and all increase hereafter, and at high level obviously than pre-impact until 6 hours after impact(P<0.05).Compared with MC group, which decrease degree more lightly and recovering quickly in Therapy group,there are obvious discrepa-ncy(P<0.01) at 6 hours after impact. left ventricular end-diastolic pressure (LVEDP) in two grou-ps all increase after SMC, and all decrease hereafter, and remained at high level until 6 hours after impact(P<0.05). Compared with MC group, which increase degree more lightly, and recovering is quickly in Therapy group, and recovering at pre-impact level until 6 hours post-impact, there are obvious discrepancy(P<0.01). Conclusion:1. In this experiment, we can establish themodel of dogs SMC at height of 85cm by diameter 5cm and 1.5kg of weight through free fall movement im-pacted surface heart.2. Myocardium glucose uptake/utilization were hindered in dogs after SMC. 3. The heart function of dogs after SMC were recovered earily by fructose-1,6-diphosphate and its therapied the dogs of SMC.
