Objective Invasion and metastasis is the main cause of gastric carcinoma mortality,and blocking the invasion and metastasis of carcinoma timely becomes the hotspot of research. This study was to investigate the expression of Syndecan-1 mRNA and HPA-1 mRNA in gastric carcinoma, and their correlation with invasion and metastasis of gastric carcinoma.Methods From May 2008 to June 2009,49 consecutive patients with gastric carcinoma admitted to our hospital for surgical treatment were included into this study. Real-time fluorescent quantitative reverse transcriptase polymerase chain reaction(RT-PCR) was used to investigate the expression of Syndecan-1 mRNA and HPA-1 mRNA in gastric carcinoma,paired adjacent gastric carcinoma (2cm from carcinoma), surgical marginal normal gastric mucosa tissues (5cm from carcinoma), and to analyze their correlation with clinic pathological characteristics of gastric carcinoma. Results The expression of HPA-1 mRNA was significantly higher in gastric carcinoma (40.56±11.75) than that in paired adjacent gastric carcinoma(18.28±11.33)and normal gastric mucosa tissues(10.80±10.20) (all the same P<0.001).The expression of HPA-1 mRNA was higher in paired adjacent gastric carcinoma than that in normal gastric mucosa tissues(P<0.05).The expression of Syndecan-1 mRNA was higher in normal gastric mucosa tissues (61.21±12.96) than that in paired adjacent gastric carcinoma (14.35±11.06)and gastric carcinoma(10.12±8.58) (all the same P<0.05).The expression of Syndecan-1 mRNA was higher in paired adjacent gastric carcinoma than that in gastric carcinoma (P<0.05).The decreased expression of Syndecan-1 mRNA and the increased expression of HPA-1 mRNA were closely associated with the degree of differentiation, depth of infiltration, lymph node metastasis, vessel metastasis and TNM staging of gastric carcinoma (all the same P<0.05).The Spearman rank correlation analysis demonstrated a significant correlation between Syndecan-1 and HPA-1 (OR=-0.405,P<0.05).Conclusion The expression of Syndecan-1 mRNA was low in gastric carcinoma,but the expression of HPA-1 mRNA was high. Meantime, the decreased expression of Syndecan-1 and the increased expression of HPA-1 can promote the invasion and metastasis of gastric carcinoma. Determination of Syndecan-1 and HPA-1 may be of value in the treatment as well as in the prognostication of gastric carcinoma.
