ObjectiveLaminectomy is one which was most commonly used for treatment of lumbar disc herniation, lumbar spinal stenosis symptom and spinal cord tumors in clinic. Failed back surgery syndrome (FBSS) refers to a condition in which the symptoms remain unchanged or worsen after surgery for lesions of the vertebrae, or interference in daily life remains. It is a syndrome of intractable chronic pain,no effective treatment has been established. The cause of this condition remains unclear, but has been variously attributed to adhesions in the epidural space after back surgery, obstruction of the peripheral nerves, and nerve root damaged. ECM abnormal depositions cause epidural fibrosis, MMPs and TIMPs could influence the formation of ECM. This article observe the effect of IL-6 on MMP-3,TIMP-1 expression in NIH3T3 Fibroblasts.Method1.Culture NIH3T3 in cell culture box with 5%CO2 and 95%O2.5th-8th cells were applied for experiment.2.Stimulate NIH3T3 Fibroblasts with IL-6(0,5,10,15,20ug/L),collect cells after 12h.3.Western bolt was applied to detect the expression of MMP-3 and TIMP-1.4.RT-PCR was applied to detect the expression of MMP-3 and TIMP-1.ResultsStimulated with different concentration of IL-6, MMP-3 expression was up-regulation with dose of IL-6,TIMP-1 expression was down-regulation with dose of IL-6.ConclusionIL-6 can affect the balance between the MMP-3 and TIMP-1 expression. Suggest it is one of the mechanisms of epidural scar formation.
