Efficacy Of Iridium Picolinate On Ovarian Cancer Cell Line SKOV-3 In Vitro And In Vivo

Objective:To study the effect of Ir on human epithelial ovarian cancer cell lines SKOV-3 in vitro and vivo by using Ir and cisplatin in different concentration to interfere in SKOV-3 in vitro and vivo in order to observe the growth state of the cells and the morphologic change of apoptosis as well as the change of cell cycle, in order to find the effective drug to treat epithelial ovary cancer patient.Methods and Materials:To cultivate SKOV-3 cell to reach logarithmic growth phase in vitro, to put the cell into the 96 pore plate and cultivate, and detect the growth curve of SKOV-3 cell, to analyze the inhibition rates of proliferation of cells by MTT after exposure to varied concentration of Ir and cisplatin for 24,48 and 96 hours. The models of solid tumor were developed by SKOV-3, calculate the relative tumor volume and observe the apoptotic rate were determined by flow cytometer.Results:The cell growth of SKOV-3 was inhibited and the effect was dependent on concentration and working time of Ir and cisplatin. There was significant difference(P<0.05 or P<0.01) to compare the OD value of cells after treated with varied concentration of Ir. The inhibition ratio on the cell growth of SKOV-3 after treated with Ir was higher than that after theated with cisplatin, and it showed time dependence. Animal experiments show that the relative tumor volume is different after treated with Ir and cisplatin. The apoptosis rate of tumor cells was higher in Ir goups than those in model groupm especially it was marked ly higher in max dose group.Conclusion:Ir plays an important role in anti-tumor effect, the cell growth of SKOV-3 inhibited and the effect was dependent on concentration and working time of Ir and cisplatin. The inhibition ratio on the cell growth of SKOV-3 after treated with Ir was higher than that after treated with cisplatin. Ir can play a positive role to improve apoptosis of tumor cell. It has the effect of disturbing cell cycle.
