To evaluate the specific effects of TCM ZHENG on pancreatic tumor growth and to uncover the potential function of "treatment based on ZHENG"("BianZhengLunZhi" in Chinese) in pancreatic cancer.We established subcutaneous tumor models of pancreatic cancer with different ZHENG including Damp heat ("Shi-Re" in Chinese), Spleen deficiency ("Pi-Xu" in Chinese), Blood Stasis ("Xue-Yu" in Chinese) in forty-eight C57/BL mice. Two days after TCM ZHENG model established, panc02 cells were injected into the right subaxillay of all mice,2×106 cells for each. The effect of each TCM syndrome on subcutaneous transplanted tumor was evaluated. Tissue samples of subcutaneous transplanted tumors from Non-ZHENG group and the ZHENG groups in which tumor growth was slowest or fastest were selected for the continuing study.The expression changes of CCR5,CXCR,CCL3,CCL4,CCL5,CXCL12 mRNA in pancreatic carcinoma subcutaneous tumor with different ZHENG were detected by RT-PCR. The protein level of CCR5, CXCR4, CD34 were observed by immunohistochemical staining. CCL3, CCL4, CCL5 which are ligands of CCR5 and SDF-1 present in subcutaneous transplanted tumor were determined using a sandwitch ELISA kit (Duoset; R&D Systems).Seventy-two C57/BL mice were randomly divided into eight groups. We duplicated models of Shi-Re, Pi-Xu and Xue-Yu. Two days after TCM ZHENG model established, panc02 cells were injected into the right subaxillay of all mice,2×106 cells for each. The response of TCM formula which was based on classic TCM theory "BianZhengLunZhi" on subcutaneous transplanted tumor with corresponding ZHENG were also studied.The rates of pancreatic cancer subcutaneous transplanted tumor models with different ZHENG are 100%.Tumor growth in Shi-Re group was slower than that in control group. Remarkably, tumors in Shi-Re group have a lower expression of CCR5, CCL4 and CCL5 than those in the control group and Pi-Xu group.Tumor growth in ZHENG groups did not decrease after administration of corresponding formula.The study indicated that during the progression of pancreatic cancer the presence of TCM ZHENG might be a protective reaction of the balance between body and the existence of tumor which was mediated by the expression of CCR5, CCL4 and CCL5;In pancreatic cancer TCM treatment, treatment determination based on syndrome differentiation almost has no effect which might be a revolution of TCM theory in cancer treatment.
