Effect Of Allitricin On The Expression Of MMP-1 And TIMP-1 In Hunan Periodontal Ligament Cells Induced By LPS

Objective To investigate the effects of allitricin and/or lipopoly-saccharide on MMP-1 and TIMP-1 expressed by human periodontal ligament cells.Methods The fifth passage of hPDLCs were cultured in media containing various concentration of allitricin and/or LPS. After 24 and 48 hours, the proteins of MMP-1 and TIMP-1 were examinded by enzymelinked immunosorbent assay(ELISA),and the ratios of MMP-1 to TIMP-1 proteins were calculated.Results 1,Compared with the control group, LPS can obviously enhance the expression of MMP-1 and such effect was time-and concentration-dependent.24 hours later, the production of TIMP-1 was inhibited.48 hours later the inhibition effect was not evident, however, the inhibition effect was obvious in 100μg/ml LPS. LPS can increase the ratio of MMP-1 and TIMP-1 significantly.2,Allitricin can induce the production of MMP-1. The time-and concentration-dependent effect were most significant in 48 hours.TIMP-1 were increased after 24 hours, while 48 hours later,9μg/ml allitricin can enhance the expression of TIMP-1.The ratio of MMP-1 and TIMP-1 had no change when irritated by 9μg/ml allitricin.It can be increased by other concentrate allitricin at the same time.3,MMP-1 were down-regulated and TIMP-1 were up-regulated by 3μg/ml allitri-cin and LPS on hPDLCs. As the time goes, 6 and 9μg/ml allitricin can induce the MMP-1 expression irritated by LPS, but have no influence on TIMP-1.3μg/ml allitricin can decrease the ratio of MMP-1 and TIMP-1.Conclusions 1,LPS can up-regulate the expression of MMP-1 while down-regulate the production of TIMP-1,and may destruct the periapical tissue by breaking the balance of MMP-1/TIMP-1.2,Allitricin can increase the expression of MMP-1,the effects were weaken as the concentration rise. It can decrease the TIMP-1 produce, but enhance the TIMP-1 production in the higher concentration as the time accumulated. Allitricin of high concentration had no influence on the ratio of MMP-1 and TIMP-1, It means that the higher concentration allitricin can balance the MMP-1/TIMP-1.3,Allitricin of light concentration can inhibit the MMP-1 expression and induce the TIMP-1 expression induced by LPS on human periodontal ligament cells at the same time. It can balance the MMP-1/TIMP-1 through influence the production of TIMP-1.
