Background:Clinical acute mushroom poisoning is a common one, which Subnigricans mushroom poisoning is more common type of mushroom poisoning. Subnigricans mushrooms, black and red mushroom also called Asia, Tan bacteria, toxic black mushroom, with the burden of sub-gate level Basidiomycetes Russula Russulaceae a poisonous wild mushrooms, is widely distributed in China, therefore, error mining eating such wild mushroom poisoning frequently. Human eating fast onset of clinical symptoms of poisoning, rapid multi-system damage caused by the body, especially liver and kidney damage is extremely clear, generally died within 72h, the fastest person can die within 6h, is the most toxic type of mushroom poisoning in a dangerous species, high mortality. Currently there is no effective antidote clinical. Ganoderma lucidum is a rare traditional Chinese medicine in China, contains a variety of active chemical components, almost all systems on the body are regulation. Clinical study found that Ganoderma lucidum decoction Subnigricans mushroom poisoning patients with good treatment, could significantly reduce Subnigricans mortality in patients with mushroom poisoning.Objective:This study animals from the point of view function and pathology confirmed that Ganoderma lucidum decoction for SD Subnigricans acute liver protective effect of mushroom poisoning, for the future of their specific detoxification mechanism further research and clinical trial basis.Methods:1. Models were:SD female rats with 20, were randomly divided into 4 groups:group control group, with normal saline; second group Ganoderma, Ganoderma lucidum decoction used; third group, the model groupâ… , using Subnigricans mushroom toxin administered modeling; the fourth group, model groupâ…¡, using Subnigricans mushroom toxin administered after model, and then Ganoderma lucidum decoction treatment.2. Study:(1) disposal group:â‘ control group:8:00 to 9:00 every morning gavage with saline 4ml;â‘¡Ganoderma:daily 8:00 am to 9:00 shrub with glossy ganoderma decoction 4ml stomach. Total treatment duration was 3 days.â‘¢model group I:the first day 8:00 am to 9:00 am with the mushroom toxin Subnigricans dubbed by 5.0g/Kg+4ml normal saline solution administered, at the same time each day, administered with saline 4ml;â‘£model group II:the first day 8:00 am to 9:00 am with Subnigricans paired mushroom toxin 5.0g/Kg+1ml saline solution orally, 2h liquid 4ml orally ganoderma later, the same day after 4ml orally ganoderma decoction with time. (2) The survival of rats and behavioral integration. End of the experiment, rats were sacrificed, heart 2ml blood serum after centrifugation of liver biochemical investigations [total bilirubin (STB), direct bilirubin (SDB), total bile acid (BA), alanine aminotransferase (ALT) and aspartate aminotransferase (AST)]; while the liver tissue formalin fixation, HE staining, and pathological changes were observed.Results:1. Survival:The model groupâ… 48h after the experiment died two, all other animals to the end of the experiment.2. Behavior:the model groupâ… rats decreased alertness, low spirit, joy to sleep, locomotor activity decreased, and the control group and comparison group of Ganoderma lucidum to reduce food intake. Biochemical changes in the liver:the model groupâ… serum STB, SDB, BA, ALT, AST was significantly higher in model groupâ…¡STB, SDB, BA, ALT, AST decreased significantly compared with model group I, and the control group but the difference between the statistics still significance. Control group and no significant difference compared Ganoderma lucidum.4. Pathological changes:(1) general visual observation:drug modules significantly enlarged liver volume, texture hardens, congestion, become darker in color. Reduced liver volume of the treatment group was not obvious, hard texture, congestion, light in color and intoxication. Texture and color of the other two were normal liver volume. (2) light microscopy:â‘ control group:Low magnification clearly seen in the liver structure, hepatocyte morphology normal, sinusoidal without expansion, hepatic lobules exist. High magnification shows normal morphology of liver cell size, abundant cytoplasm, nuclear size and normal, clear nucleoli, no signs of necrosis.â‘¡Ganoderma:Low magnification clearly seen in the liver structure, hepatocyte morphology rules, the same size, lobular structure exists. High magnification shows normal morphology of liver celstructure l size, abundant cytoplasm, nuclear size and normal, clear nucleoli, no signs of necrosis.â‘¢model group I:Low magnification significant structure seen in liver damage, congestive heart, liver cells were irregular, inconsistent size, swelling, condensation nuclei, sinusoidal expansion, congestion and destruction of lobular structure. High magnification shows irregular liver cells, size inconsistency, cloudy swelling, granular cytoplasm poisoning, nucleus condensation, part of the nuclear membrane dissolves or disappears, showing necrosis, sinusoidal expansion, congestion.â‘£model groupâ…¡:Low magnification can see a clear organizational structure of the liver, liver cells irregular shape, size, more consistent, mild swelling, sinusoidal slightly expanding, slightly congestive. High magnification shown in the liver cells were essentially normal, the cytoplasm was slightly turbid, cloudy swelling, nucleolus, hepatic sinusoid slightly expanded, a little congestion, no expansion of interlobular vein, bile duct non-proliferation.Conclusions:1. Subnigricans mushrooms on the SD rats with acute liver damage effects.2. Ganoderma lucidum decoction of acute mushroom poisoning Subnigricans liver function in rats has a protective effect.3. Ganoderma lucidum decoction of acute mushroom poisoning Subnigricans liver cell damage in rats has a protective effect.
