The Decreased Expression Of Cdc42 On Peripheral Lymphocytes In Acute Ischemic Stroke And Its Relationship With Golgi Apparatus

Objective:Peripheral lymphocytes adhere to the endothelium and infiltrate into infarcted tissue after acute ischemic stroke. Cdc42, a small GTPase, is responsible for mediating adhesion and migration of lymphocytes. It is known that Golgi apparatus is involved in the metabolism of Cdc42. Little is known about the changes of Cdc42 expression on peripheral lymphocytes after acute cerebral ischemia. Here, our present study was designed to investigate the expression of Cdc42 on peripheral lymphoctes in actue stage of ischemic stroke, further explore the potential roles of Golgi apparatus in the mechanisms of Cdc42 expression.Methods:38 patients with acute cerebral ischemia in the Department of Neurology who was within 72 hours after onset were recruited as ischemic stroke group, and 38 healthy people were recruited as control group. By immunofluorescence, the subcellular locations of Cdc42 in peripheral lymphocytes were exhibted. The expressions of Cdc42 and TGN46 in peripheral lymphocytes were respectively detected by immune-cytochemistry. Furthermore, the correlation between TGN46 and Cdc42 expressed in peripheral lymphocytes were studied.Results:1. Cdc42 was localized in the cytoplasm of peripheral lymphocytes. 2. In ischemic patients, the percentage of Cdc42 positive lymphocytes was less than that of control group (P<0.05).3. The percentage of TGN46 positive lymphocytes from peripheral blood of the patients with ischemic stroke was also decreased in comparison with control group.4. In ischemic group, the expressions of Cdc42 and TGN46 on peripheral lymphocytes were positively correlated (r=0.92, P<0.05).Conclusions:1. The expression of Cdc42 in peripheral lymphocytes was decreased in patients after acute cerebral ischemia.2. TGN46 expressed in peripheral lymphocytes was down-regulated after acute cerebral ischemia, which inicated the impaired functions of Golgi apparatus.3. Down-regulation of Cdc42 in peripheral lymphocytes was related to the impaired function of Golgi apparatus in acute stage of ischemic stroke.
