OBJECTIVES:Slow transit constipation (STC)In Clinical it refers to constipation, colonic transit time and cellulose, laxatives characterized by poor response to treatment, accompanied by abdominal pain, bloating, nausea and other symptoms, and to remove the basis of systemic disease and pelvic floor muscle dysfunction of chronic constipation , colonic motor function by the disturbance caused by the extension of colonic transit time and colon characterized by decreased power stubborn constipation.Foreign scholars have found through the study of animaland human specimens that,Progesterone concentration of estrogen in women and the abnormal slow transit constipation have a very close relationship.To investigate the abnormal expression of G protein and progesterone in slow transit constipation SD rat, and to provide the theoretical basis for the slow transit constipation SD rat. For slow transit constipation treatment clinics to provide effective clinical thinking.SUBJECT:SD female rats of clean gradeMETHODS:Establish slow transit constipation model in SD rats by Loperamide.An experimental model of chronic slow transit constipation rat. After the success of the model, extract STC rat blood, and serum progesterone within the content, then extract rat colon STC by cleavage digest and by using Western blot analysis, use G protein quantitative to determine the content of the test box.RESULTS:The expression of progesterone in normal SD rat: (25.23±6.81)ng/ml.The expression of progesterone in slow transit constipation SD rat: (5.1±3.98)ng/ml. The expression of G protein and progesterone in slow transit constipation SD rat has significant deviation compared with the normal rat. Expression of G protein in normal SD rat Gαs:(84.6±8.4)% and Gαq:(159.4±7.2)% .Expression of G protein in slow transit constipation SD rat Gαs:(140.4±20.9)% and Gαq: (90.6±4.7)%.CONCLUSIONS:1.STC rats progesterone concentrations than normal rats significantly decreased the concentration of progesterone;2.STC rats than in normal rats may be caused by down Gαq of contractile Gαs and up-regulation.
