Abstract/Summary:
Objective To investigate the effect of Intermedin on oxidative stress in ischemia/reperfusion injury in rat kidney.Methods A totle of thirty male Wistar rats were randomly divided into five groups (control group, IRI group, empty plasmid group, ADM group and IMD group). after remove of right kidney ultrasound plasmid transfected into the kidney, after 1 week artery clipping 45min, preparation 24h reperfusion IRI model. semi-quantitative analysis of renal pathology injury, detection of renal tissue SOD, MPO, caspase-3 activity, MDA content, Immunohistochenistry text ICAM-1, ET-1 and P-selection expression. TUNEL detection of apoptosis of renal tissue.Results 1 the changes of the renal morphological, the most severe groups happened in the IRI group, IMD pretreatment significantly decreased the degree of renal injury.2 IRI group significantly decreased SOD activity, increased MDA production; increased MPO activity, increased ICAM-1 and P-selection expression; increased ET-1 expression; increased the activity of caspase-3, increased the renal tubular epithelial cells wither death. IMD pretreatment significantly decreased SOD activity, decreased the MDA production; decreased MPO activity, decreased the ICAM-1 and P-selection expression; decreased ET-1 expression; decreased the activity of caspase-3, decreased the renal tubular epithelial cells wither death. their is no difference between empty plasmid group and IRI group.Conclusion IMD pretreatment protects against renal I/R injury. The mechanism maybe at least partlly related to the clearance of oxygen free radicals, decreased lipid peroxidation, inflammatory cell infiltration and cell apoptosis, thereby reducing oxidative stress bring the ROS.
