1,Expression and significance of the growth-factor independence 1 in LeukemiaObjective To study the expression and clinical significance of the grouth-factor independence 1(Gfi1) in leukemia. Methods Bone marrow mononuclear cells were obtained from 65 leukemia patients including 24 acute myeloid leukemia(AML),18 chronic myelogenous leukemia (CML),6 acute lymphoblastic leukemia (ALL),17 blast crisis of Chronic Myelogenous Leukemia and 13 IDA patients as control..The relative expression of Gfi1 was examined by taqman quantitative real-time reverse transcription polymerase chain reaction. Results Expression of Gfi1 in leukemia was higher than that of control group and the difference was statistically significant(P<0.01). Expression of Gfi1 in CML was higher than that in AML,ALL,CML-BP and .the difference was significant (P<0.01). Expression of Gfi1 in lymphocytic blast crisis of CML was higher than that of myelocytic blast crisis of CML.Conclusion Gfi1 played an important role in leukemia ,especialy in CML and its progression. The high lever expression of Gfi1 may be participate in the development of lymphoma.2,Expresion and significance of Gfi1 in Leukemia CellsObjective To detect the expression of Gfi1 in leukemia cell lines and its role in the development and differentiation of myeloid blood cells. Methods The expression levels of Gfi1 in leukemia cell lines—NB4,K562 were induced with PMA or retinoic acid by reverse transcription-polymerase chain reaction(RT-PCR) and Western blot when they were in logarithmic phase. Results. During ATRA-induced granulocytic differentiation of NB4 and K562 cells, the expression of Gfi1 mRNA is higher than that with PMA. The Gfi1 protin levels has the same trend during differentiation of the two leukemia cells with ATRA. During PMA-induced monocytic differentiation of K562 cells, The Gfi1 protin levels is higher than that with ATRA but mRNA levels drop.Conclusion During granulocytic differentiation of HSC Gfi1 might play an important role .Gfi1 has the same role during monocytic differentiation of HSC.
