Objective:To explore the affection of NGB expression in retina after eyeball contusion by oxygen and to study the injury repair mechanisms and treatment of eyeball contusion in high altitude by investigating the regulation of the neuroglobin (neuroglobin, NGB)expression level in rabbit retina in the plateau environment after eyeball contusion.Methods:30 (n= 60) Beijing Chinchilla rabbits fed a plateau at an altitude of 3784m environment. Made these rabbits anesthesia by ketamine on 25mg/kg intramuscular injection. Before striked by 0.33kg of steel ball at central corneal once, these rabbits need to be fixed their head vertically under the lm height PVC pipe and observated their retinal condition. These injuried eyeball rabbits were randomly divided into two groups,15 for the plateau oxygen group, the other 15 for the plateau of non-oxygen group. The oxygen group was a experimental group. The non-oxygen group was the control group. The oxygen group oxygen 10 hours everyday by 35%, Measured oxygen saturation at 8:00 am and 18:00 pm. The non-oxygen group were done the same measure. Two groups of animal models were killed after wound 12 h,1d,3d,7d and 14d by gas injected in ear vein.3 (n= 6) rabbits were sacrificed at each time point. Then removed eye balls immediatelly and made windows on the scleral. After that fixed them in the 40g/L paraformaldehyde. The eye ball cut ringly at 5mm of the scleral margin after two weeks, including retina and optic nerve. The organization did routine dehydration, transparent, embedded in paraffin, serial cut 6 sections of 3μm thick. These pieces were mounted upon the glass slides handed with APES (Poly-Lai acid) and dried more than 6h,. Then did the HE method and immunohistochemical ABC method. Analysis of one way variance would be used for the date of mean gray±S of NGB in the retina compared different time points in one group and t-test would be used the date of mean gray of NGB in the retina in order to compared the different of the two groups in the same time point. P<0.05 as statistically significant.Results:1. The rule of expression of NGB in altitude hypoxia environment on retinal is that the NGB expression was gradually increased at 12h, Id,3d and 7d, in which NGB expression increased significantly at 3d and 7d, but weakened at 14d. The expression of NGB of oxygen group was gradually increased at 12h, 1d,3d and 7d, but significantly decreased at 14d.2. Our experiments show that the expression of NGB in the retina of oxygen group is lower than that in non-oxygen group. There was no statistical difference of NGB expression between two group at 12h, 1d (P12h= 0.648> 0.05, P1d= 0.500> 0.05), The expression of NGB in oxygen group is weaker than those of non-oxygen group at 3d and 7d with statistical significance, in which the differences are significant at 7d (P= 0.008<0.01), There was no statistical significant of NGB expression between two group at 14d (P= 0.642> 0.05).Conclusion:1, The rule of expression of NGB oxygen group in altitude hypoxia environment on retinal is simily to the non- oxygen group. 2, Oxygen would affect the expression of NGB in rabbit retina after eye contusion in high altitude and the expression of NGB with oxygen threatment reduced varylly.3, Released the retina injur and enhanced the reparation of retina by improving the expression of NGB through adopting low dense of oxygen treatment.
