Object:BALB/c mice planted subcutaneously with Renca cell are used to establish renal cell carcinoma models, and study the drug including IL-2 and COX-2 inhibitor NS-398 effect to the ectopic tumor growth, and the changes of CD4+CD25+Foxp3+ regulatory T cells (Treg) in peripheral tumor-bearing mice blood, spleen, and changes in local tumor. COX-2 inhibitors increase of renal function and mechanism of immune therapy, COX-2 inhibitors for the adjuvant treatment of kidney cancer and provide experimental basis for clinical application.Methods:1. Babl/c mice planted subcutaneously with Renca cell are used to estabish renal cell carcinoma models.2. Tumor-bearing mice are Randomly divided into A, B, C and D four groups, different groups take different delivery methods, A group is IL-2 group, B groupis NS-398 group, C group is IL-2+NS-398 group, D group is for the PBS control group.3. After administration of 14 days and 28 days of extraction of peripheral blood tumor-bearing mice in each group, with changes in Treg by flow cytometry.4. Excised tumor and spleen of tumor-bearing mice, using immunohistochemistry to detect mouse spleen and tumor local changes in TregResults:1. Tumor in mice by HE staining plant pathology, a large number of abnormal cells seen under a microscope and see the mitotic phase, tumor cells were round, prismatic or irregular in shape, cells larger, less cytoplasm, nuclear large, deeply stained, cells were arranged in disorder, showing solid-like structure, interstitial less, showing invasive growth, the structure around the non-coated confirmed Babl/c mice were found growing ectopic kidney tumor characteristics, you can use Babl/c mice from the renal cell carcinoma cell line (Renca) establish a mouse model of ectopic kidney2. Babl/c mice with ectopic renal cell carcinoma treated with different drugs in significant changes in tumor volume, A by IL-2 treatment group within 10 days of tumor volume growth is still slow,16 days after the tumor began to decrease, reducing the rate lower than large; B group only NS-398, cancer is still growing, but slower growth than the control group; C group for the combined treatment group, tumor reduction occurs in the sixth day, then continued to the end of the experiment, by the multiple t inspection, and other groups with significant difference (P<0.05).3. COX-2 inhibitors can lower the expression of Treg in blood, tumor and spleen and IL-2 combined with COX-2 inhibitors creat a synergistic effect (P<0.05).Conclusion:1.Use Babl/c mice to establish the source of Renca cells Babl/c mouse model of ectopic kidney can be used as tumor model of murine renal cell carcinoma.2. COX-2 inhibitor combined with IL-2 could significantly inhibit tumor growth in mice ectopic kidney.3. COX-2 inhibitors can lower blood cell, tumor and spleen in the expression of Treg, Treg suppress the reduction in tumor growth is one of the mechanisms.4. IL-2 combined use of COX-2 inhibitors can be used as Babl/c mice with ectopic kidney cancer immunotherapy adjuvant.
