| Phytic acid is a kind of natural and organic phosphorus compounds, widely found in cereals, pulses and oil plant seeds. Because of its low toxicty,strong chelation of metals and significant anti-oxidation,the applications of phytic acid in food, environmental, medical and chemical industry are concerned by more and more people. Therefore, how to make full use of plant resources to get phytic acid has strong market potential in our country. Meanwhile, the phytic acid in the diet on mineral absorption inhibition usually shows dose-response,and low concentrations of phytic acid doesn't affect the body's absorption of these elements.So it has the positive significance for guiding people with a reasonable diet and selecting the appropriate processing methods of cooking to avoid the adverse effects on absorption that phytic acid may cause to know the relationship between phytic acid and people's health,the content in food,the intake of different groups of people and the relationship between phytic acid and mineral.In the meanwhile, it also can provide references for the further researches on such as removing-phytic acid processing and phytic acid enhancement in food project.In this paper, the extraction of phytic acid, the parameters of purifying phytic acid with resin,the antioxidant activity of phytic acid in vivo and vitro,and the antibacterial activity of phytic acid were studied with skimmed apple seed as raw material.(1)The extraction of phytic acid showed:_hydrochloric acid was used as the solvent for phytic acid extraction by optimizing,and the extraction time,extraction temperature,liquid ratio and acid concentration,which had greater impact on the extraction rate of phytic acid,were chosen as the single factor for response surface experiment,and the best extraction parameters of phytic acid were: temperature 35℃,time 0.75h, liquid ratio of 17:1,solvent concentration 0.4 mol/L.(2)The phytic acid purification by anion resin experiment showed:LX-300 anion resin was chosen to purify the phytic acid,and the phytic acid could be completely eluted by 0.7mol/mL NaCl liquor;the elution peak had favorable symmetry,and the recovery rate is 90.2%;the cellected liquid was low-tension water bath till paste was formed,and the concentrated phytic acid concentration was 49.45%.(3)The phytic acid antioxidant tests in vitro showed:the clear effects of phytic acid to DPPH free radical,·OH free radical and O2-·free radical was better than VC,and the IC50 were separately 9.31 mg/mL,2.54 mg/mL and 9.28 mg/mL.The phytic acid had better reducing power than Vc did,and the IC50 which inhibited the lipids peroxidation was 3.24 mg/mL.(4)The antioxidant effect of phytic acid in mice experiment showed:phytic acid could improve the activity of the SOD,CAT and GSH-PX and remarkably reduce the content of MDA in mice's serum and hepatic tissue,and the T-AOC of mice was improved in the same time.(5)The bacteriostasis tests of phytic acidshowed:the phytic acid and the phytate sodium both had obvious bacteriostatic effect to staphylococcus aureus,colibacillus,bacillus subtilis and bacillus cereus with worse bacteriostatic effect to thermophilic bacteria;to the same becteria, the phytic acid had better bacteriostatic effect than the phytate sodium did. |
