Objection:To investigate the effect on weight loss and blood lipid in the animal body of four strains of lactobacillus separated from normal intestine of human which had been proved to have the cholesterol-lowering function in vitro.It was aimed to provide the base on selecting the probiotics which were favorable to human.Also observnig the effect on the liver lipid deposition of the fat rat model and discussing the possible mechanism.Methods:70 SD rats for experiment were randomly divided into 7 groups.Group K was normally fed,While Group C(08 GW13),D(08 HJ3),E(MM1-16),F (9-41-A)were fed with high-fat diet and various probiotics strains at the same time.Group I is the model group fed with high-fat diet.Group H is the positive control group intervented with atorvastatin. In the process of feeding, the weight and length of the rats was measured in every morning of the eighth day.6 weeks later,weight and lee's index were measured before executed,then all rats were executed after taking blood.The effect of various lactobacillus strains on blood lipid were determined with serum total cholesterol (TC) and triglycerides (TG), low density lipoprotein cholesterol (LDL)-c), high-density lipoprotein cholesterol (HDL-c).Then the effect on the liver was observed by detecting the liver weight,liver index,iver lipid and the malondialdehyde (MDA) content and superoxide dismutase (SOD) vitality of the livers.Liver pathological sectionswere also needed to be observated Under the microscope.Result:The body weight decreased significantly in group D,F.The body weight dropped from 348.71±17.09 in model group to 330.00±8.56(D) and 325.57±10.04(F) respectively.The lee's score(in group D,E and F) was decreased in compare with the model groupⅠ(P<0.05). Liver weight and liver index of each probiotic group with model group were no obviously difference.Besides the group D,TG level of other groups were decreased obviously(P<0.05).TG level of positive control group(H) was the lowest.The TCH level in group C,F was decreased significantly(P<0.05).The effect on the LDL-c and HDL-c was little (P>0.05).TG and TCH in livers of probiotics groups were all lower than the model group's(P<0.05).Then the malondialdehyde (MDA) content in liver of group F,H was lower than that of the model group. The superoxide dismutase (SOD)vitality in liver of group C,F and H was higher than that of the model group I.Among them,positive control group(H) had the lowest MDA content and the highest SOD vitality,the difference compared to group I was of significance (P<0.05).Conclusion:1.Lactobacillus 9-41-A strain separated by our laboratory had the obvious effect on lowing weight decreasing the blood lipid and inhibiting liver lipid peroxidation.2.Lactobacillus 08 HJ3 strain have discreased body weight in rats fed with high-fat diet, Lactobacillus 08 GW13 strain and MM 1-16 strain can reduce the serum lipid and the lipidoses in liver.
