OBJECTIVES:The diabetes is harm to human health seriously, and the incidence is increasing year by year. There is a lack of endogenous insuli in the patients of type 1 and type 2 diabetic. At present, the clinic treatment can not meet the reconstruction needs of pancreaticβcell function. In currently found, the MafA gene can induce insulin secretion in vitro, and has great significance to the treatment to the diabetes. In this study,we investigate the MafA gene therapy on blood glucose and insulin secretion of the diabetes rats.SUBJECT:MafA gene, Wistar rats with type 2 diabetes.METHODS:The MafA plasmid was transformed into E.coli DH5α, firstly we select clones, and then amplificat the plasmid. Secondly, we use the low-dose plasmid extraction kit to extract the plasmid.Induce the modle of diabetic wistar rats by the STZ, such as the ratio of the mixture of MafA and liposomes by injecting to the Portal vein of the diabetic rats, and the observe the change of the blood glucose of the diabetic rats.RESULTS:The blood glucose of treatment group by 13.6mm/L~14.8mm/L decreased to 20.6mm/L~22.8mm/L, not only the blood glucose desend about 5mmol/L and the plasma insulin levels were significantly elevated, but aslo they can maintain about two weeks; After the treatment,the blood glucose below was the untreated group(group DM)(P<0.05) ; After the treatment, the plasma insulin levels were significantly higher than the untreated group(group DM) (P<0.05); The liver of the groupA can detect the expression of the MafA mRNA, and the groupDM can not detect the expression. The result of the immunohistochemical analysis revealed that the insulin was found in the liver of the treatment group, and the insulin was not expression in the liver of the group DM.CONCLUSIONS:The therapy of injecting the ratio of the mixture of MafA and liposomes through the Portal can decrease the blood glucose of the diabetic rats.
