Traditional Chinese medicine is Chinese experiences of long-term medical practice and wisdom, it makes Made a great Contribution to the Chinese people's survive and multiply during the thousands of years. Salvia miltiorrhiza as a traditional medicine has been used for a long time in China. It has important medicinal value. Purify the active ingredient of Salvia can improve the efficacy and advance the standardization of Chinese medicines.The liposolubility and water-soluble substances have different chemical properties, so we try to use the different methods to separation and make it more efficient. In order to meet the industrial production process, we also development and optimizate the extraction methods of tanshinoneⅡA and salvianolic acid B.Mesolow preparation chromatographic process has been used in Salvia miltiorrhiza separation. Compared with the Traditional ordinary presure column chromatography. This method can shorten the separation time, improve the production efficiency. The experiments use the 500 mesh silica gel as the immobile phase, use acetidin and ligroin(9:1) as mobile phase. After the separation and recrystallization, we can get four compounds which have high purity. They are dihydrotanshinoneⅠ, tanshinoneⅠ, cryptotanshinone, tanshinoneⅡA.Separating the water-solubility component by kiese gel has a low efficiency, so we ues the HSCCC to Separate. After a lot of experimental optimizations we set up a solvent system with petroleum ether-acetic ether-methanol-water-formic acid. The best method is:rotation 1800r·mL-1, flow rate:1.0 mL·mL-1, temperature:30℃in experimental conditions. The material obtained is light green powder, after recrystallization, we get colorless powerder. Analyze by HPLC, the purity of the salvianolic acid B is greater than 98.00%. After analyzing, it is salvianolic acid B.TanshinoneⅡA and salvianolic acid B are the main liposolubility and water-soluble substances in Salvia miltiorrhiza. We often make the extraction contents of the two components as the standard of liposolubility and water-soluble substances contents. We use ultrasound extraction in this study, make the standard curves of tanshinoneⅡA and salvianolic acid B by external standard method and measure the contents by HPLC. The best extraction technology has been identified by the results of the single factor experiments and orthogonality experiments. The results show that the best method is methanol concentration 40%, solvent-material ratio 13 and ultrasounding time 30 min. Ultrasound extraction, with advantages of quickness, convenience and efficiency, may cut down the costs in industrial production.
