Backgrounds:The present study were aimed to detect the changes of the fatty liver targets in serum and liver sample. Moreover, experiments were designed to elucidate the potential underlying protective mechanisms and optimal safe ischemic time of preconditioning on hepatic I/R injury in steatotic livers.Methods:The study were designed to make the fatty liver model by feeding a high-fat diet. The changes of targets in serum and in liver sample were evaluated for the different ischemic time of preconditioning liver. Histological analyses of liver were performed,and evaluation of hepatic fatty infiltration were performed by sudan III stain.Results:1. Fatty liver modal was successful made. Almost 50% hepatocyte have fatty infiltrated,and some up to 100% hepatocyte. And with no fatty liver appearance occurring in rats in ordinary diet group.2. The preconditioning consisting of 15 minutes of ischemia followed by 10 minutes of reperfusion was performed (15 min -10 min IPC), the extent and number of necrosis areas at 24 hours after hepatic reperfusion was markedly increased in fatty livers with respect to I/R. By contrast, The histological study in normal liver showed moderate.3. The level of serum and MDA, MPO in liver found in 5 or 8 min -10 min preconditioning group were lower than the other preconditioning groups in fatty liver, while the SOD activity lever was significantly higher than the others (p < 0.05). The serum level of NO in IPC group has significant higher than the corresponding I/R group (p < 0.05). In fatty liver group, the level of NO 5 or 8 min - 10 min IPC group was significantly higher than the others (p < 0.05).Conclusion:1. Liver with fat infiltration can aggravated ischemia-reperfusion injury in rats.2. Ischemia preconditioning can also increase the tolerance of fatty liver induced by high-fat diet to hepatic ischemia-reperfusion injury in rats. The protective role of ischemia preconditioning were distinct in different ischemic time of preconditioning, and the 5 ~8 min - 10 min IPC was superior to the others.
