[Objective] To explore the abnormity of bone marrow microenvironment of psoriasis patients by studying the level of cytokine secreted by bone marrow mesenchymal stem cells and the level of cytokine receptor expressed by bone marrow mononuclear cells of psoriasis patients.[Methods]â‘ Bone marrow mononuclear cells of patients and control were separated by density gradient centrifugating. The bone marrow mesenchymal stem cells were separated and cultured with the method of adherent. The phenotypes of the cells were identified by flow cytometry.â‘¡The level of IL-8,IL-3,GM-CSF and HGF in supernatant liquid were assayed by ELISA kits.â‘¢The expression of IL-3Ra and HGFR were assayed by flow cytometry.â‘£Psoriasis area and severity of the rash using PASI score method, the level of cytokines and its relationship with disease severity using pearson correlation analysis.[Results]â‘ Flow cytometry showed that the bone marrow mesenchymal stem cells expressed surface antigens CD44,CD71 and CD34,CD45,HLA-DR expression is negative.â‘¡The level of IL-8,IL-3 and HGF secreted by bone marrow mesenchymal stem cells of the patients was significantly lower than that of control (P<0.05), however, the level of GM-CSF was no significant difference between the two groups (P>0.05).â‘¢The expression of IL-3Ra and HGFR on bone marrow mononuclear cells of the patients was significantly lower than that of control (P<0.05)â‘£The cytokine level of psoriasis patients and PASI score had no correlation.[Conclusion]â‘ The psoriatic bone marrow microenvironment is abnormal. The abnormal microenvironment may be play a important role in the pathogenesis of psoriasis.â‘¡The cytokine level of psoriasis patients and disease severity had no correlation.
