OBJECTIVE: To investigate the expression and correlation of P-Akt and NFкB in cervical carcinoma tissues. To explore whether P-Akt and NFкB is useful in the occurrence and development of the cervical cancer and the Correlation with Human Papilloma Virus(HPV). So new ideas are provided for cervical cancer diagnosis,treatment and prevention.METHODS: The expression of P-Akt and NFкB were detected by immunohistochemical SP technique in 26 cases of cervical cancer tisuues, 18 cases of cervical intraepithelial neoplasia tisuues (CINⅠn=5,CINⅡn=3,CINⅢn=10) and 19 cases of chronic cervicitis tisuues.The different expression of P-Akt and NFкB were compared in cervical cancer different pathological type (cervical squamous cell carcinoma, cervical adenocarcinoma) and the different pathological grading (high, medium , poorly differentiated) and different clinical stage (FIGO I to IV) .The relationship between P-Akt and NFкB respectively with HPV infection in cervical cancer were correlation analyzed.RESULTS:The positive expression rate of P-Akt in chronic cervicitis tisuues,CIN and cervical cancer tisuues was 21.05%,66.67%,92.31% respectively.There was no obvious difference among the express of P-Akt in cervical cancer in different pathological type and in pathological grading (P>0.05) ,and no obvious difference among different clinical stage(P>0.05).The positive expression rate of NFкB in chronic cervicitis tisuues,CIN and cervical cancer tisuues was 10.52%,72.22%,96.15% respectively; There was no statistically significant difference among the groups(P>0.05)indifferent pathological type(P>0.05),and there was no obvious difference among in different pathological grading and in different clinical stage(P>0.05). There was obviously positive correlation between P-Akt and NFкB expression rate and the degree of disease (r=0.998, P <0.05) Cervical cancer and CIN cases are 44.The positive expression rate of P-Akt was 87.55% in the 32 cases of HPV-DNA positive of the 44 cases ,and the positive expression rate of P-Akt was only 16.70% in the 12 cases of HPV-DNA negativetive of the 44 cases . The positive expression rate of NFкB was obviously higher in the HPV-DNA positive than in the HPV-DNA negativetive. There was statistically significant difference among the groups(P <0.05).CONCLUSIONS: The positive expression rate of P-Akt and NFкB is closely related with cervical disease extent,and closely related with HPV infection in cervical cancer. suggesting that P-Akt and NFкB more probably play an important role in the occurrence and development of the cervical cancer.
