Objective: ginseng saponins is the main pharmacological composition of ginseng which is the traditional Chinese medicine.We want to know whether it have or not the protective effects for lung tissue against acute lung injury in rats and discuss its possible mechanismMethods:40 Sprague Dawley(SD) rats only were randomly divided into four groups: control group, model group, ginseng saponins (GS) group, dexamethasone group. Each drug was injected from the tail intravenous. after the beginning of the Experiment , controls and model group were injected 1ml Physiological saline. GS group were injected GS solution by 100mg/kg, dexamethasone group were injected dexamethasone liquid by 5mg/kg. After An hour, the control group was injected 1ml NS, Model group, GS group and dexamethasone group will were injected endotoxin by 5mg/kg. After 6 hours, we would observe the general situation of rats and measure the blood gas analysis from Abdominal aortic. Then we removed the left lung to weigh the wet weight, dry weight and Calculate lung coefficient (wet/dry weight ratio). Then we took the right lung to observe generally with the naked eye and did Pathological slices to abserve the changes by 200 times light-microscopy. At Last,we measured the content of SP-A protein and SP-B protein from Bronchoalveolar Lavage Fluid by Ellisa medthod.Results:1.40rats were without loss and death in this experiment, we would analysis the results of all rats.2.General situation: after 6 hours from medicine injection had been finished, the ventilatory frequency of control group was: 56-64 times a minutes and the rats had no abnormal signs . The ventilatory frequency of remaining each rat were: model group: 121-134 times a minute, ginseng saponins groups: 95-106 times a minute, dexamethasone groups: 83-91 times a minute. All rats appeared the differment degree of respiratory distress and limb cyanosis. 3.By observing generally Lung macroscopic:The shape of control group rats were smooth and no abnormalities, The color of Model group were dark red , which large bleeder and infarcts were found . Dexamethasone groups: slight swelling and a few gel-foam cubes.Saponins group:hyperemia ,edema and visible gel-foam cubes. By the use of the 200 times Optical microscope : the structure of the control group was clear and no special changes;Model group: hyperaemia and infiltrates was visible from the tiny blood vessels. a number of liquid oozy , a lot of inflammatory cells infiltration and multifocal of atelectasis was visible in the Pulmonary intersitital and alveolar space. The exudation of the Ginseng saponins group was lighter than model groups, small inflammatory hyperaemia of lung tissue in tiny blood vessels and cell infiltrates in interstitial and alveolar space in Dex group. 5. Arterial blood gas: compared with controls, thePaO2 and PaO2 / FiO2 from rest groups decreased obviously (P < 0.01). compared with the damage group, saponins group and dexamethasone group were relativly taller (P < 0.01), compared with the saponin Groups, dex group was slightly higher (P < 0.05) .Compared with the control groups, the expression of SP-A protein and SP-B protein from other three groups were all lower (P < 0.01). Compared with the model group, the expression of two kinds of protein was relativly taller (P < 0.01),saponins group and dexamethasone group of expression Group compared with the saponin group, the expression of two kinds of protein from dexamethasone group was slightly higher (P < 0.05).Conclusions:1.Ginseng saponins could relieve respiratory distress degree and pulmonary edema ,improve ventilation function for acute lung injury in rats. 2.Ginseng saponins could treat acute lung injury in rats by inhibiting the reduction of the content of the SP - A protein and SP - B protein from lung tissue in the rats.
