Background and Objective: The hepatitis B virus (HBV)is a major pathogen threatening human health.A complex interaction between pathogen and host genetics determine both susceptibility to particular microorganisms and the course of infection.Recently much interests have been focused on susceptibility to HIV,HBV,and SARS.In the post-genomic era, SNPs have become critical elements for properly analyzing gene association.Our purpose is to investigate the relationship between the genetic polymorphisms of the Fas ligand,which is an apoptosis-associated protein, and severe hepatitis B in Han Chinese.Methods: A case-control analysis was adopted in this study, in which 233 cases of chronic asymptomatic HBV carrier,68 cases of severe hepatitis B,102 cases of HBV related cirrhosis,112 cases of HBV related primary hepatocellular carcinoma,and 100 cases of innate immunization were recruited from seven hospitals from June,2008 to May, 2009.FasL-844T/C polymorphism for each sample was examined by TaqMan real time fluorescent quantitative polymerase chain reaction(RT-qPCR).A adjusted odds ratios(ORs)and 95% confidence intervals(CIs)were calculated using the logistic regression model. Results: Binary logistic regression analysis indicated that FasL-844CC of chronic asymptomatic HBV carriers is significantly linked to a higher risk for severe hepatitis B after adjusting for gender and age (OR=4.729,P=0.043).In other groups,there were no statistic significance(P>0.05).Conclusions: Chronic asymptomatic HBV carriers harboring FasL-844CC may have greater susceptibility to severe hepatitis B.We ought to pay more attention to this populations.
